摘要
目的研究神经纤毛蛋白1(neuropilin 1,Nrp1)在卵巢癌中的作用,探讨其作为治疗靶点的可行性。方法克隆Nrp1基因,采用慢病毒包装体系包装过表达Nrp1的重组慢病毒,感染卵巢癌SKOV3细胞,嘌呤霉素筛选稳定高表达Nrp1的SKOV3细胞,通过细胞计数检测其增殖能力的改变。结果成功包装了过表达Nrp1重组慢病毒,筛选得到了稳定高表达Nrp1的SKOV3细胞,其增殖能力显著高于对照细胞(t>t_(0.05(4)),P<0.05)。结论 Nrp1促进卵巢癌SKOV3细胞增殖,可能在卵巢癌发生发展过程中起促进作用。
Objective To study the function of neuropilin 1 (Nrp1) in ovarian cancer and the possibility of Nrp1 as therapeutic target. Methods Nrp 1 gene was cloned, and Nrp 1 overexpressing recombinant lentivirus was packed.After infection of SKOV3 ovarian cancer cells, puromycin selection generated the stable cell clones overexpressing Nrp 1.Cell number counting assay was performed to detect the cell proliferation. Results Nrp 1 overexpressing recombinant lentivirus was successfully packed. The stable SKOV3 cell clones overexpressing Nrp 1 was selected, and it had more powerful proliferation ability than control cells (t〉t0.05(4), P〈0.05).Conclusion Nrp1 promotes the proliferation of SKOV3 cells, and may stimulate the progression of ovarian cancer.
