摘要
目的通过比较坑道噪声性听力损失易感人群和非易感人群血清蛋白指纹图谱的差异,寻找显著性表达的差异蛋白,为筛选更适合从事特殊兵种作业的官兵提供参考。方法根据噪声性听力损失易感人群分布情况,选择每个兵种高度易感组20人为模型建立组,非易感组18人为对照组,共38人,收集其血清样本。应用磁珠联合基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption/ionization time of flight mass spectrometry,MALDI—TOF—Ms)对两组样本进行差异蛋白研究,筛选出显著性差异的血清蛋白,并应用遗传算法建立诊断模型。结果噪声性听力损失易感人群组与对照组之间共检测出55个差异血清多肽峰,其中有统计学意义的差异峰仅有2个,分子量分别为1083.57Da和1061.68Da(P〈0.05)。以这两个差异蛋白建立数据模型,结果提示该模型诊断敏感度为54.24%,特异度为91.67%。结论噪声性听力损失易感人群组与对照组之间存在差异显著的血清蛋白,应用遗传算法建立血清差异蛋白的诊断模型,可为筛选更适合从事特殊兵种作业的官兵提供参考。
Objective To compare the differences in serum protein fingerprints between population susceptible to noise-induced hearing loss and non-susceptible controls, to find out significant differences in protein expressions and to provide reference for the selection of more suitable soldiers for the Special Forces. Methods According to the distribution of population susceptible to noise-induced hearing loss, 20 cases susceptible to noise-induced hearing loss were selected as the model group and 18 cases as the non-susceptible control group. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) technology was used to analyze the serum of the two groups and to detect the serum protein with decreased expression. The genetic algorithm was applied to establish the diagnostic model. Results 55 protein peaks were detected in the two groups. There were significant differences in the 2 proteins with mass-to-charge ratio (m/z) at 1083.57 Da and 1061.68 Da between model group and control group (P〈0.05). The 2 proteins were chosen to establish the diagnostic model. The sensitivity and specificity were 54.24% and 91.67% respectively. Conclusion There are significant differences in the expressions of serum proteins between model group and control group. The diagnostic model of serum differential proteins with genetic algorithm can be used for screening more suitable soldiers for the Special Forces.
