摘要
为研究制何首乌水提液对小鼠成骨细胞MC3T3-E1的影响,将不同浓度的制何首乌水提液作用于MC3T3-E1细胞,采用噻唑蓝(MTT)法、碱性磷酸酶(ALP)活力检测试剂盒和实时荧光定量PCR法对细胞增殖、ALP蛋白分泌及骨保护素(OPG)和核因子κB受体活化因子配体(RANKL)mRNA表达水平进行检测。结果显示,药物作用2d和3d后,各浓度制何首乌水提液均可促进MC3T3-E1细胞增殖;药物作用6d后,各浓度制何首乌水提液均可促进MC3T3-E1细胞分泌ALP;药物作用1d后,较高浓度制何首乌水提液可上调MC3T3-E1细胞OPG/RANKL mRNA表达比。表明制何首乌水提液具有促进成骨细胞增殖、分化和上调成骨细胞OPG/RANKL mRNA表达比的作用。
To study the effects of Radix Polygoni Multiflori Preparata water extracts on mouse osteoblasts (MC3T3-E1 cells),the cells were treated with different concentrations of Radix Polygoni Multiflori Preparata water extracts,then the cell proliferation,the alkaline phosphatase(ALP)secretion and the mR-NA expression of OPG and RANKL were measured by MTT,ALP assay kit and quantitative real-time PCR assays.The results indicated that,after 2 d and 3 d,the proliferation of MC3T3-E1 cells treated with different concentrations of water extracts was promoted;after 6 d,the ALP secretion of MC3T3-E1 cells treated with different concentrations of water extracts was up-regulated;after 1 d,the OPG/RANKL mR-NA expression ratio of MC3T3-E1 cells treated with high concentration of water extracts was up-regula-ted.In conclusion,Radix Polygoni Multiflori Preparata water extracts could promote the proliferation and differentiation of osteoblasts while up-regulate the OPG/RANKL mRNA expression ratio of osteoblasts.
