乙型肝炎病毒感染相关肝病外周血单核细胞中miRNA139的表达变化及临床意义 被引量:2

摘要 目的:探讨乙型肝炎病毒感染相关肝病中miRNA139的表达情况及其临床意义。方法:应用实时荧光定量PCR方法检测慢性乙型肝炎(CHB)患者(20例)、慢性乙肝肝硬化(LC)患者(20例)、原发性肝癌(HCC)患者(20例)及正常对照(NC)(20例)组外周血单核细胞中miRNA139的表达水平,结合临床指标进行分析。结果:与NC组比较,CHB组和LC组患者外周血中miRNA139的表达水平下调不明显,没有显著差异(P〉0.05);与NC组、LC组和CHB组比较,HCC组患者外周血中miRNA139的表达水平明显下调,差异具有统计学意义(P<0.01)。HCC患者外周血中miRNA139的表达与AFP呈负相关。结论:HCC组患者外周血中miRNA139的表达水平明显下降,提示miRNA139的下调可能与HCC的发生有关。 Objective:To investigate the expressions and clinical significance of microRNA(miRNA)139 in the peripheral blood mononuclear cells(PBMCs)of patients with Hepatitis B. Methods:The levels of miRNA139 in PBMCs of patients with CHB, LC,HCC and normal controls were detected by fluorescence real-time quantitative PCR method and combined with clinical correlation analysis.Results:There was no statistical difference in the miRNAl39 expression in PBMCs between patients with CHB,LC and normal controls .The levels of miRNA139 in PBMCs of patients with HCC were lower than that of normal controls and patients with CHB,LC(P〈0.01).Negative correlation was found between the miRNA 139 and AFP in HCC patients,but no correlation with HBV-DNA or ALT. Conclusions:The reduction of miRNA 139 expression related to the incidence of HCC. Detection of miRNA 139 might help in diagnosis and prediction of prognosis of HCC.
