摘要
目的观察胡桃醌对He La细胞凋亡的影响并探讨其机制。方法用四甲基偶氮唑蓝比色法[3-(4,5-dimethy-2-thiazoly)-2,5-diphenyl-2-tet-razoliumbromide,MTT]观察胡桃醌对He La细胞生长抑制情况;Hoechst 33342荧光染色法检测He La细胞凋亡;免疫荧光染色法观察凋亡蛋白active caspase-3和内质网应激标志物PDI的表达;免疫印记法(Western blotting)检测Grp78、PDI和CHOP表达水平。结果胡桃醌对He La细胞的生长抑制作用呈现剂量依赖关系,差异有统计学意义(P<0.05);胡桃醌可以使He La细胞active caspase-3表达增强,并伴有内质网应激标志蛋白Grp78、PDI表达水平增高,差异有统计学意义(P<0.05),还能使内质网应激途径凋亡调控蛋白CHOP表达水平增高,差异有统计学意义(P<0.05)。结论胡桃醌能够抑制He La细胞增殖,并通过内质网应激途径诱导其凋亡。
Objective : To observe the effect of juglone on apoptosis of HeLa cells and explore the mechanism. Methods : MTr assay was used to observe the inhibitory effect of juglone on growth of HeLa cells ; Hoechst 33342 fluorescent staining was used to detect apoptosis of HeLa cells; immunofluorescence staining was used to observe the expressions of active caspase-3 and PDI protein; Western blot was used to detect the expression levels of Grp78, PDI, and CHOP. Results : The inhibitory effect of juglone on growth of HeLa cells showed a dose-dependent manner, there was statistically significant difference (P〈0. 05 ) ; juglone could increase the expression of active caspase-3, Grp78, PDI, and CHOP (P〈0. 05) . Conclusion : Juglone can inhibit the proliferation of HeLa cells and induce apoptosis of HeLa cells by endoplasmic reticulum stress.
