摘要
本文采用响应面法优化湖南永州生姜茎叶总黄酮的提取条件。在单因素试验基础上,选择乙醇浓度、提取温度、提取时间为影响因子,应用Box-Benhnken中心组合法进行3因素3水平试验设计,以生姜茎叶总黄酮得率为响应值,进行响应面分析,并研究了生姜茎叶总黄酮的体外抗氧化活性。结果表明,生姜茎叶总黄酮的最佳提取条件为乙醇浓度70%、提取温度76℃、提取时间95 min,在此条件下,生姜茎叶总黄酮的得率可达到15.42±0.09 mg/g。同时建立了乙醇溶液提取生姜茎叶总黄酮的二次数学模型,对目标产物提取具有良好的预测作用。生姜茎叶总黄酮体外清除羟基自由基能力的IC50值为0.501 mg/m L,其总还原能力和清除羟自由基能力均高于相同浓度的2,6-二叔丁基对甲酚(BHT),但低于L-抗坏血酸(Vc)。
Response surface methodology (RSM) was employed to optimize the extraction conditions of total flavonoids from Zingiber officinalee Roscoe stem and leaves collected from Yongzhou (Hunan province). Based on single-factor tests, Box-Benhnkcn center-composite experiment was carried out with three factors,including alcohol concentration, ex- traction temperature and extraction time. RSM was used to determine the effect of prime factors on the yield of fla- vonoids. Besides,the antioxidant activity of flavonoids from Z. o.~winale stem and leaves was evaluated. Results showed that the optimal extraction conditions were ethanol concentration 70% ,extraction temperature 76 ~C and extraction time 95 min. Under these conditions,the extraction yield of total flavonoids was 15.42 ± 0.09 mg/g, which was close to the predicted value. Thus,the fitted quadratic regression model was valid. The ICso of the obtained total flavonoids against hydroxyl radicals were 0.501 mg/mL. The reducing ability and scavenging impact of the radicals of flavonoids from Z. o t^tcinale stem and leaves were more effective than those of 2,6-di-tert-butyl-4-methyl phenol (BHT) at the same con- centration, but lower than vitamin C.
