间变少枝胶质细胞瘤血管内皮细胞1p/19q缺失与生存期的关系 被引量:3

Relationship between 1p/19q deletion of vascular endothelial cells in anaplastic oligodendroglioma and survival period

摘要 目的探讨间变少枝胶质细胞瘤(AO)血管内皮细胞1p/19q缺失情况,并分析其与生存期的关系。方法收集首都医科大学附属北京天坛医院2009年1月至2010年1月具有完整随访资料的原发性AO标本30例,其中男16例,女14例;年龄23~61岁,平均45.9岁。免疫荧光技术定位血管内皮细胞,荧光原位杂交法(FISH)检测1p/19q缺失状态。配对t检验统计血管内皮细胞1p/19q缺失率,生存分析法评估其与生存期的关系。结果AO血管内皮细胞的检测信号1p36缺失率显著高于其参照信号1q25(t=8.823,P〈0.001),检测信号19q13缺失率显著高于其参照信号19p13(t=8.316,P〈0.001)。生存分析结果显示:血管内皮细胞1p/19q共缺失的患者中位无进展生存期(PFS)及总体生存期(0s)均显著优于血管内皮细胞1p/19q未缺失的患者(PFS:X^2=16.199,P〈0.01;OS:X2=8.666,P〈0.01)。Cox回归分析结果表明血管内皮细胞1p/19q共缺失是独立预后因子(PFS:HR=0.063,95%CI:0.013—0.313,P=0.001;OS:HR=0.145,95%CI:0.310~0.678,P=0.014)。结论AO血管内皮细胞存在1p/19q缺失现象。血管内皮细胞1p/19q共缺失提示预后良好。 Objectives To investigate the lp/19q deletion status of vascular endothelial ceils in anaplastic oligodendroglioma (AO) and to analyze its relationship with survival period. Methods A total of 30 primary AO specimens with complete follow-up data from Beijing Tiantan Hospital, Capital Medical University were collected between January 2009 and January 2010. There were 14 females and 16 males, and their mean age was 45.9 years ( range 23 to 61 ). Immunofluorescence technique was used to locate vascular endothelial cells. Fluorescence in situ hybridization (FISH) was used to detect the status of lp/19q deletion. The paired t test was used to count lp/19q deletion rate of vascular endothelial cells. Survival analysis method was used to assess its relationship with survival period. Results The detection signal l p36 deletion rate of AO vascular endothelial cells was significantly higher than its reference signal 1 q25 (t = 8. 823, P 〈 0. 001 ) ; the detection signal 19q13 deletion rate was signifcantly higher than its reference signal 19p13 ( t = 8. 316, P 〈 0. 001 ). The results of survival analysis showed that both the median progression-free survival (PFS) and overall survival (OS) of the patients with lp/19q co-deleted of vascular endothelial cells were significantly better than those without lp/19q deletion of vascular endothelial cells (PFS : X2 = 16. 199,P 〈 0.01 ; OS : X2 = 8. 666, P 〈 0.01 ). The result of Cox regression analysis showed that lp/19q co-deletion of vascular endothelial cells was an independent prognostic factor (PFS: HR = 0.063, 95% CI0.013-0.313, P=0.001; OS: HR=0.145, 95% C10.310-0.678, P=0.014). Conclusions AO endothelial cells have a lp/19q deletion phenomenon, lp/19q co-deletion of vascular endothelial cells indicates that the prognosis will be good.
