Expression of Vitamin D receptor in the myocardium of mice with viral myocarditis

摘要 目的探讨维生素D受体(VDR)在病毒性心肌炎(VMC)小鼠心肌中的表达动态变化和作用。方法选取120只4周龄雄性BALB/c小鼠,随机分为对照组(n=40)和实验组(n=80),实验组小鼠腹腔注射柯萨奇B3病毒制备VMC模型,对照组小鼠腹腔注射等量的DMEM液作为替代。分别在注射3、7、14和28 d后各处死15只实验组小鼠和10只对照组小鼠,取小鼠心脏标本。用免疫组织化学法检测小鼠心肌组织VDR的表达动态变化,并用苏木精-伊红染色观察小鼠心肌的病理变化。结果实验组小鼠接种病毒后VDR表达显著升高,并在第7天表达水平达到高峰,然后逐渐下降,在第28天仍处于较高水平(P<0.01);在第3、7、14和28天,实验组小鼠VDR表达水平均高于对照组,差异均有统计学意义(P<0.01);实验组小鼠心肌病理评分变化趋势与VDR表达变化趋势一致;实验组小鼠心肌VDR表达水平与其心肌病理变化呈正相关(P<0.01)。结论 VDR可能参与了VMC发病的炎性免疫过程。 Objective To investigate the dynamic expression and role of vitamin D receptor (VDR) in the myocardium of mice with viral myocarditis (VMC). Methods One hundred and twenty 4-week-old male BALB/c mice were selected and assigned into control (n=40) and experimental groups (n=80). The mice in the experimental group were injected intraperitoneally with Coxsackievirus B3 to establish the model of VMC, while the mice in the control group were injected intraperitoneally with an equal volume of DMEM solution. Fifteen mice in the experimental group and ten mice in the control group were sacriifced at 3, 7, 14, or 28 days after injection, and the myocardial specimens were obtained. The dynamic expression of VDR in the myocardium was determined by the immunohistochemical technique. The pathological changes in the myocardium were examined using hematoxylin and eosin staining. Results In the experimental group, the mice had significantly increased expression of VDR after virus injection (P〈0.01);the expression of VDR reached the peak at 7 days after injection, and then declined gradually; the expression of VDR remained high at 28 days after injection. At 3, 7, 14, and 28 days after injection, the expression of VDR in the experimental group was significantly higher than that in the control group (P〈0.01). Moreover, in the experimental group, the changes in the pathological score of the myocardium were in accordance with the changes in the expression of VDR;the expression level of VDR in the myocardium was positively correlated with the pathological changes in the myocardium in the experimental group (P〈0.01). Conclusions VDR may be involved in the inlfammatory-immune process in the pathogenesis of VMC.
