摘要
目的探讨建立最佳2型糖尿病合并脂肪肝大鼠模型方案。方法用不同方案给予链脲佐菌素、烟酰胺及高脂饲料干预大鼠8周后,制备脂肪肝动物模型。尾静脉取血,测定空腹血糖(FPG),全自动生化分析仪检测总胆固醇(TC)、三酰甘油(TG)、肝和血清谷丙转氨酶(ALT)、谷草转氨酶(AST);放免法检测空腹胰岛素(FINS),计算肝指数及胰岛素抵抗指数;光镜下观察肝组织改变。结果先注射链脲佐菌素50 mg·kg-1、高脂饲料喂养后,大鼠血糖、血脂、肝功能有显著的变化,组织病理学观察肝细胞出现脂变。结论先注射STZ后高脂饲料喂养可以成功复制糖尿病并脂肪肝动物模型,且符合人类此病的发病特点。
Objective To explore establishment method of ideal rats model with diabetes complicated with fatty liver. Methods SD rats were induced by high -fat, streptozocin and nicoti-namide. The animal model with diabetes complicated fatty liver induced by streptozocin and nieoti- namide was established. Eight weeks later, the serum of total cholesterol ( TC ), triglyceride ( TG), glutamic - pyruvic transaminase ( ALT), glu- tamic oxalacetic transaminase (AST) , fasting glucose (FPG) , fasting in- sulin (FINS) were measured. And the pathological changes of liver tissues were observed. Results There were markedly changes in blood glucose, blood lipid and transaminases and similar histopathology to fatty liver in rats with STZ 50 mg kg-1 and high - fat. Compared with the other groups, this group is more typical and more consistent with diabetes and fatty liver pathologic changes. Conclusion The way of injecting STZ adding high fat - feed can estabilish successfully the animal model of dia- betes with fatty liver. And this model was also in line with human charac- teristics of this desease.
