强志怡神胶囊的大孔树脂纯化工艺优选 被引量:1

Optimization of Purification Process of Qiangzhi Yishen Capsules with Macroporous Absorptive Resin

摘要 目的:优选强志怡神胶囊的大孔树脂纯化工艺。方法:以细叶远志皂苷、芍药苷、甘草酸、甘草苷的比吸附量和洗脱率为指标筛选大孔树脂型号。采用单因素试验考察上样液质量浓度、径高比、洗脱剂体积等因素对强志怡神胶囊大孔树脂纯化工艺的影响。结果:选用HPD-100型大孔树脂,最佳纯化工艺为上样液质量浓度0.1 g·m L-1,树脂-生药量(3∶1),树脂径高比1∶7.5,上样速度3 BV·h-1,加4 BV水洗除杂,继加70%乙醇5 BV洗脱,洗脱速度3 BV·h-1。细叶远志皂苷、芍药苷、甘草酸、甘草苷的转移率分别为78.55%,88.62%,82.47%,83.15%,得膏率7.52%,干燥时间0.5 h。结论:该工艺能有效去除远志、白芍、甘草水提液中的淀粉、糖类等杂质,所得纯化物易于制剂成型,可作为强志怡神胶囊的纯化工艺。 Objective: To optimize purification process of Qiangzhi Yishen capsules with macroporous resin. Method: Absorption ratio and elution rate of tenuifolin, paeoniflorin, glycyrrhizie acid and liquiritin were taken as indexes to screen out types of macroporous resins. Effects of the concentration of solution, diameterheight ratio and eluent volume on purification process of Qiangzhi Yishen capsules were investigated by single factor tests. Result: HPD-100 maeroporous resin was selected, optimum purification process was as following: the mass concentration of sample loading solution 0. 1 g·mL^-1, resin-material drug (3:1 ) , diameter-height ratio of resin 1:7.5 ,sample loading speed 3 BV ·h^-1, decontaminated with 4 BV of water, and then eluted with 5 BV of 70% ethanol, elution speed of 3 BV·h^-1. Transport rates of tenuifolin, paeoniflorin, glycyrrhizic acid and liquiritin were 78.55%, 88.62%, 82.47%, 83.15%, with rate of extract of 7.52% and drying time of 0.5 h. Conclusion: This process can effectively remove starch, saccharides and other impurities in water extract, these purified substances is easy to use in preparations.
