摘要
目的建立HPLC-MS/MS法测定川楝子中川楝素的方法。方法采用XTerra C18色谱柱(150mm×2.1mm,5μm),以乙腈-0.2mL·L^-1甲酸溶液(31∶69)为流动相,采用多反应监测(MRM)模式进行质谱测定。结果川楝素检出限(LOD)为0.17ng,定量限(LOQ)为0.51ng;川楝素质量浓度在0.05-5.00μg·mL^-1范围内,与峰面积线性关系良好,r=0.999 9;精密度良好,平均回收率为98.0%,RSD为1.5%。结论该方法具有较好的专属性和灵敏度,能够简便、快速地测定川楝子药材中川楝素的含量。
Objective To establish a method of HPLC‐MS/MS for detecting toosendanin (TSN ) in Toosendan Fructus . Methods The analytes were isolated on a XTerra C18 column (150 mm × 2 .1 mm ,5 μm) by isocratic elution with acetonitrile‐mL · L^ -1formic acid(31∶ 69)as the mobile phase ,and detected by MS/MS in a multiple reaction monitoring (MRM) mode . Results LOD of TSN was 0 .17 ng ,and LOQ of TSN was 0 .51 ng ;The linear range of TSN concentration was 0 .05‐5 .00μg · mL^ -1 (r=0 .999 9);The average recoveries were 98 .0% with RSD 1 .5% .Conclusion The method is simple and rapid with high sensitivity and good reproducibility ,and it could be used to determine the contents of TSN in Toosendan Fructus .
