摘要
目的探讨二维应变技术(2D-SE)定量评价法舒地尔对糖尿病合并动脉硬化大鼠左心室功能影响的价值及机制。方法将27只Wistar大鼠随机分为对照组(Cont组,9只)、模型组(Mod组,9只)和法舒地尔组(Fas组,9只)。Cont组普通饲料喂养;其他2组给予尾静脉注射链脲佐菌素及高脂饮食5周,建立糖尿病合并动脉硬化模型,之后Fas组给予法舒地尔治疗4周。观测左心室整体径向应变(GRS)、圆周应变(GCS)及血管性血友病因子(v WF);分析应变参数与v WF的相关性。结果 Mod组大鼠v WF水平高于Cont组及Fas组(P<0.05),而Fas组与Cont组比较差异无统计学意义(P>0.05)。Mod组与Cont组及Fas组相比GRS、GCS明显降低(P<0.05),Fas组GRS、GCS较Cont组降低(P<0.05)。GRS、GCS与v WF呈负相关(P<0.05)。结论法舒地尔对糖尿病合并动脉硬化大鼠左心室功能的影响与改善内皮功能相关,2D-SE评价糖尿病合并动脉硬化大鼠的左心室功能是一种有效的方法。
Objective To establish a diabetic and atherosclerosis rat models,so as to discuss the mechanism of the effect of Fasudil on left ventricle function using two-dimensional strain echocardiography(2D-SE). Methods Among the 27 Wistar rats selected for the study,9 were selected as control group(Cont group),the other 18 were used to establish diabetic and atherosclerosis models. Then the modeling group was equally divided into Model group(Mod group,n =9)and Fasudil group(Fas group,n =9). The Fas group was given Fasudil with the dose of 10 mg·kg-1·d-1for 4weeks. The global radial strain(GRS),global circumferential strain(GCS)and blood levels of von Willebrand factor(v WF)were measured. The correlation between strain parameter and v WF were analyzed. Results The v WF level was significantly higher in Mod group compared to Cont group and Fas group(P〈0.05);however,there were no significance difference between Cont group and Fas group(P〈0.05). The GRS and GCS were significantly decreased in Mod group compared to Cont group and Fas group(P〈0.05). Moreover,the GRS and GCS in Fas group were significantly reduced compared to Cont group(P〈0.05). There were negative correlation among v WF,GRS and GCS. Conclusion Fasudil could influence the heart function of diabetic and atherosclerotic rats by improving vascular endothelial function. 2D-SE method is useful in the evaluation of left ventricle function in diabetic and atherosclerotic rats.
