摘要
目的:药对防风-乌梅抑制肥大细胞系P815细胞分泌Th2类细胞因子的作用及机制,为该药对临床应用提供科学依据。方法:胰蛋白酶刺激P815细胞方法建立肥大细胞脱颗粒模型,用ELISA法观察药对防风-乌梅对肥大细胞分泌Th2类细胞因子IL-4、IL-13及相关信号通路蛋白Akt、ERK的影响。结果:药对防风-乌梅在体外可阻断肥大细胞IL-4的分泌,并抑制信号通路蛋白Akt、ERK的表达,与细胞模型组比较有统计学差异(P<0.05或P<0.01)。结论:药对防风-乌梅抗哮喘机制可能与该药对通过抑制肥大细胞ERK和AKT蛋白,阻断肥大细胞分泌IL-4有关。
Objective: To research the effect in vitro of Fangfeng-Wumei compatibility to Th2-type cytokines produced by mast cell line P815 and the related mechanism, and provide an exact basis of the compatibility for clinical application. Methods: We estab- lished a mast cell degranulation mouse model by stimulating mouse mast cell line P815 with trysin in vitro, and then observed the level of IL-4 and IL-13, and signaling pathway-related proteins including Akt and ERK, affected by Fangfeng-Wumei, using ELISA. Results : Fangfeng-Wumei compatibility in vitro significantly inhibited the secretion of IL-4 by mast cells, and suppressed the expression of signaling pathway-related proteins, involving Akt and ERK, in comparison to cell model group ( P 〈 O. 05 or P 〈 0. 01 ). Conclusion: By inhibiting the expression of AKT and ERK in mast cells, Fangfeng-Wumei compatibility suppressed IL-4 se- cretion of mast ceils and thus displayed anti-allergic activation.
