腺苷蛋氨酸配伍美洛昔康治疗膝骨关节炎的实验研究 被引量:11

Protective effect of S-adenosy-L-methionine combined with meloxicam on knee osteoarthritis in rats

摘要 目的:研究腺苷蛋氨酸(SAMe)配伍美洛昔康(MLX)对大鼠膝骨关节炎(OA)的治疗作用,并探讨其相关作用机制。方法:单侧膝关节腔内注射碘乙酸钠(MIA)诱导大鼠OA模型,评价不同时间点大鼠平均关节炎指数(MAI),光镜观察大鼠膝关节软骨和滑膜的组织病理学改变,ELISA法检测血清和关节液中一氧化氮(NO)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-a(TNFα)含量及血清中软骨寡聚基质蛋白(COMP)、基质金属蛋白酶-3(MMP-3)含量。结果:SAMe,MLX及复方均可不同程度减轻OA大鼠膝关节炎症症状;改善膝关节软骨和滑膜的病变;降低大鼠血清和关节液中IL-1β,TNFα和NO含量;降低大鼠血清中COMP和MMP-3含量。结论:SAMe和MLX都能减缓OA大鼠的病理进程,两者合用可不同程度提高疗效,其作用机制可能与血清和关节液中的炎症细胞因子减少以及血清中COMP和MMP-3失调有关。 Objective: To investigate the effect of S-adenosy-L-methionine (SAMe) combined with meloxi- cam (MLX) on knee osteoarthritis (OA) in rats, and the possible mechanism. Methods: OA was induced by in- jection of sodium mono-iodoacetate (MIA) in unilateral knee joint. The mean arthritis index (MAI) was evaluated at several time-points, and the histopathology of knee joint cartilage and synovial was obserwed by light microscopy. Cartilage oligomeric matrixprotein (COMP), matrix metalloproteinase-3 (MMP-3), nitric oxide (NO), cytokine interleukin-1β (IL-1β) and tumor necrosis factor-or (TNF-α) in serum or synovial fluid were detected by ELISA. Results : SAMe, MLX and their combination obviously reduced the degree of inflammatory symptoms in knee joint, improved the lesions in knee articular cartilage and synovium, reduced the activity of COMP and MMP-3 in serum, and reduced the levels of IL-1β, TNF-α and NO in serum and synovial fluid. Conclusion: SAMe and MLX slow the pathological process in OA rats, and their combination further improves the protective effect, which may be as- sociated with reduction of COMP and MMP-3 in serum as well as inflammation factors in serum and synovial fluid.
