Reelin基因缺失对小鼠小脑浦肯野细胞迁移与极化的影响 被引量:1

The Effect of ReelinDeletion on Migration and Polarization of Cerebellar Purkinje Cells in Mice

摘要 利用免疫荧光双标、尼氏染色技术对不同发育阶段Reelin基因缺失的reeler小鼠与野生型小鼠小脑浦肯野细胞的发生及迁移进行形态学观察,同时比较和测量其极性改变的角度.结果显示,reeler小鼠小脑皮质在发育过程中片层化发生紊乱,浦肯野细胞未按正确位置迁移,其胞体直径和极性化角度改变显著,与野生型小鼠比较,差异具有统计学意义(P<0.01).以上结果说明,Reelin基因缺失使小脑浦肯野细胞极性改变,迁移受阻,提示Reelin蛋白在小脑皮质片层化形成的过程中发挥了至关重要的作用. Using double immunofluorescence and Nissl staining technology, we observed the neurogenesis and migration of the purkinje cells and measured their polarity change perspective in reeler mice and wild type mice. Results show that all layers of the cerebeUar cortex in reeler mouse are disorder severity, and the migration of purkinje cells was blocked. The diameter of cell body and the polar angle changes greatly in reeler mouse purkinje cells, with statistically significant difference (P 〈0. 01). The above results show that Reelin deletion can affect the neurogenesis and migration of cerebellar purkinie cell, suggesting Reelin protein play a crucial role in the formation of cerebellar cortical lamination.
