KCNE2基因rs10854373多态位点与冠心病遗传易感性的关系 被引量:1

摘要 目的观察KCNE2基因单核苷酸多态位点rs10854373的基因型及等位基因频率分布,探讨KCNE2基因rs10854373多态性与广东地区汉族人群冠心病遗传易感性的关系。方法随机选择893例冠心病患者(冠心病组)和816例健康对照个体(对照组),采用直接测序的方法,对KCNE2基因rs10854373多态位点进行分型,采用非条件逻辑回归分析统计该多态位点与冠心病遗传易感的相关性。结果 CC、CT、TT在冠心病组分布频率分别是59.9%、34.8%、5.3%,在对照组中分别为58.9%、37.1%、4.0%,两组间基因型频率分布差异无统计学意义(2=2.809,P=0.246);非条件逻辑回归分析发现,携带T变异等位基因与冠心病遗传易感性无明显相关性(OR=1.02,95%CI为0.86~1.19,P=0.812)。校正吸烟、糖尿病、高血压、高脂血症后,两组等位基因频率分布差异仍无统计学意义(OR=0.95,95%CI为0.79~1.15,P=0.627)。此外,对吸烟、高血压、糖尿病和高脂血症等危险因素进行分层分析显示,KCNE2基因rs10854373多态位点与冠心病的易感性亦无相关性。结论 KCNE2基因rs10854373多态位点与冠心病遗传易感性无相关性。 Objective To investigate the association between the single nucleotide polymorphism (SNP) of KC- NE2, rs10854373, and the genetic susceptibility of coronary artery disease (CAD) in the Han ethnical group in Guang- dong Province. Methods The genotyping of the SNP was conducted using direct sequencing in both 893 CAD patients and 816 non - CAD controls. Association between the polymorphism of KCNE2 rs10854373 and the risk of CAD was eval- uated through unconditional logistic regression analysis. Results The frequencies of CC, CT, TF in CAD group were 59. 9%, 34. 8%, 5.3%, respectively; and 58.9%, 37.1%, 4.0%, respectively, in the control group. No significant difference was observed in frequency of KCNE2 rs10854373 geuotypes between cases and controls ( x^2 = 2. 809, P = 0. 246). Unconditional logistic regression analysis did not demonstrate significantly increased risk of GAD for those carry- ing T variant allele (OR = 1.02, 95% CI 0. 86 - 1.19, P = 0. 812). And even after adjustment of hypertension, diabe- tes, hyperlipidemia and smoking, no significant difference was shown in frequency of KCNE2 rsi0854373 genotypes be- tween cases and controls (OR = 0. 95, 95% CI 0. 79 - 1.15, P = 0. 627). Moreover, there was no significant association between this polymorphism and increased CAD risk in subgroup analysis factoring in hypertension, diabetes, hyperlipi- demia and smoking. Conclusion Genetic polymorphism of KCNE2 rs10854373 is not associated with coronary artery dis- ease in the Han ethnical group in Guangdong Province.
