摘要
目的 探讨脂联素对脓毒症大鼠肝组织NLRP3炎性小体及炎症因子表达的影响。方法清洁级7~8周龄雄性Wistar大鼠30只,随机均分为三组(n=10):假手术组(S组)、模型组(M组)和脂联素组(APN组)。采用盲肠结扎穿孔(CLP)法建立脓毒症模型。S组剖腹后仅分离盲肠远端,不结扎穿孔。APN组于CLP前12h腹腔注射基因重组脂联素6mg/kg。CLP后24h处死,留取血样本后取肝组织标本。活性氧(ROS)的含量采用流式细胞技术测定、酶联免疫吸附法测定IL-1β水平、Western Blot检测NLRP3及caspase-1含量。结果 与S组比较,M组和APN组大鼠血清IL-1β水平明显升高、肝组织ROS、NLRP3及caspase-1含量明显增加(P〈0.05);与M组比较,APN组大鼠血清IL-1β水平明显降低,肝组织ROS、NLRP3及caspase-1含量明显减少(P〈0.05)。结论脂联素可改善脓毒症大鼠炎性反应,其机制可能与降低ROS含量、抑制NLRP3炎性小体活性有关。
Objective To investigate the effect of adiponectin on the expression of NLRP3 in- flammasome and inflammatory factors in liver tissue of septic rats. Methods Thirty healthy male Wistar rats, aged 7-8 weeks were randomly divided into 3 groups (n = 10) : group sham operation (group S), group model (group M) and group adiponectin (group APN). Septic model was per- formed by cecum ligation and puncture (CLP). Group S only took the operation of isolated distal cae- cum, without ligation perforation. In group APN, gene recombinant adiponectin 6 mg/kg was injec- ted intraperitoneally at 12 h before CLP. Rats were been executed 24 h after CLP, liver tissue were harvested after obtained the blood samples. Flow cytometry was used for determining the level of Re- active oxygen species (ROS), ELISA was performed for the level of IL-1β, Western Blot was used for the levels of NLRP3 and caspase-1. Results Compared with group S, the levels of serum L-1β and ROS, NLRP3, caspase-1 content in liver tissues were significantly increased in group M and group APN (P〈0. 05); Compared with group M, the levels of serum L-1β and ROS, NLRP3, caspase-1 content in liver tissues were significantly decreased in group APN (P〈0. 05). Conclusion Adiponec- tin might regulate immune response in septic rats by reducing the level of ROS and inhibiting the NL- RP3 inflammasome activity.
