柱前衍生高效液相色谱法分析纸张中的单糖

摘要 建立了3-氨基-9-乙基咔唑(AEC)柱前衍生高效液相色谱法测定纸张水解液中单糖的方法:以3-氨基-9-乙基咔唑为柱前衍生试剂,Thermo C18色谱柱为分离柱,乙腈和乙酸铵水溶液为流动相,在波长245 nm处,用高效液相色谱分析纸张中的单糖。结果表明:确定了衍生化试剂和硼氢化钠的最佳用量,反应时间为70 min,反应温度为70℃。该方法具有操作简单、灵敏度和准确度高等优点,为纸张中单糖的检验提供了一种实用的方法。 A new method of analysis of monosaccharides in paper was established by HPLC with pre-column derivatization using 3-amino-9-ethylcarbazole. Monosaccharides in paper were derivatized using 3-amino-9-ethylcarbazole, separated by reverse Thermo C18 column, using acetonitrile and ammonium acetate aqueous solution as mobile phase and detected by HPLC under the wavelength of 245 nm. The optimum amount of derivatizing agent and sodium borohydride were determined, results showed that the reaction time was 70 min, the reaction temperature was 70 ℃. The method was simple, sensitivity and accuracy, and provided a practical method for testing monosaccharides in paper.
