实验性脾虚大鼠肠道黏膜味蛋白Gαgust基因及蛋白表达及补脾方干预研究 被引量:3

Study on the expression of gene and protein of gut taste protein Gαgust in experimental rats with spleen deficiency and the intervention of the tonifying spleen prescription

摘要 目的:观察脾虚模型大鼠肠黏膜味蛋白(Gαgust)表达情况以及健脾方药的干预作用,探讨肠道味觉关键信号分子Gαgust异常表达与脾虚证的发病相关性。方法:采用大黄脾虚模型,并设肾虚模型对照,通过QPCR法、Western blot法、免疫组化法对脾虚治疗组、脾虚自然恢复组、脾虚模型组、肾虚对照组及正常对照组Gαgust进行检测。结果:脾虚模型组Gαgust表达与正常对照组、脾虚自然恢复组相比下降(P<0.05),尤其与肾虚组比较也有显著性差异(P<0.05),经健脾治疗后脾虚治疗组Gαgust表达比脾虚自然恢复组、脾虚模型组、肾虚对照组表达上升。结论:肠道味觉关键信号分子Gαgust异常表达与脾虚证的发病密切相关,而健脾方药对其有一定的调节作用。 Objective: To discuss the relationship between the abnormal expression of the key signal molecule of gut taste and the occurrence of spleen deficiency by observing the expression of gut taste protein Gttgust of model rats with spleen deficiency and the intervention of the tonifying spleen prescription. Methods: Using spleen deficient model caused by rhubarb and establishing kidney deficiency group, we detected the key signal molecule of gut Gαgust of spleen deficiency treatment group, spleen deficiency natural recovery group, spleen deficiency control group, kidney deficiency group and normal control group by QPCR method, WB method and Immunohistochemical method. Results: It shows that compared with the spleen deficiency control group and the spleen deficiency natural recovery group the expression of the key signal molecule of taste Go, gust of spleen deficiency treatment group decreased (P〈0.05), especially decreased significantly (P〈0.05), when compared with the kidney deficiency group. The expression of gene and protein of Gctgust the kidney deficiency group has no significant difference with normal control group. After the treatment of tonifying spleen, the expression of Gαgust of the spleen deficiency treatment group increased, compared with the spleen deficiency control group, the spleen deficiency natural recovery group and the kidney deficiency group. Conclusion: The abnormal expression of Gαgust, which is the key signal molecule of gut taste related closely to the occurrence of spleen deficiency and the tonifying spleen prescription has specific regulating effect on it.
