摘要
【目的】探讨哮喘小鼠气道和肺组织中血管内皮细胞生长因子(vascular endothelial growth factor, VEGF)及抑瘤素 M(oncostatin M,OSM)的表达及布地奈德的干预作用。【方法】45只健康6~8周龄雌性清洁级 BALB/C 小鼠随机分为:哮喘模型组(A 组)、布地奈德干预组(B 组)和空白对照组(C 组),每组15只。观察各组小鼠气道及肺组织的病理变化,免疫组化及 RT-PCR 检测 VEGF 及 OSM 的蛋白和 mRNA 表达。【结果】VEGF 和 OSM 的蛋白及 mRNA 在 A 组的表达显著高于 B 组和 C 组,且差异有显著性(P <0.05),A组 OSM 蛋白和 VEGF 蛋白的表达呈正相关(r =0.41,P <0.05)。【结论】①VEGF 和 OSM 在哮喘小鼠的气道和肺组织中表达增高,布地奈德干预可使其表达降低。②VEGF 和 OSM 参与哮喘的气道重塑过程,两者呈直线相关。③布地奈德干预可以通过减少 VEGF 和 OSM 的表达影响哮喘气道重塑。
[Objective]To explore the expressions of vascular endothelial growth factor (VEGF)and on-costatin M (OSM)in airways and lungs of asthmatic mice and examine the interventional roles of budesonide.[Methods]A total of 45 BALB/c specific-pathogen free female mice were randomly divided into control,asth-ma and budesonide intervention groups (n = 1 5 each).The pathological changes of airways and lung tissues were observed in all mice.The protein expressions of VEGF and OSM were detected by immunohistochemis-try.And the mRNA levels of VEGF and OSM in lung tissues were determined by reverse transcription-poly-merase chain reaction (RT-PCR).[Results]The protein and mRNA expression of VEGF and OSM in airway and lungs of asthma group significantly higher than those in control and budesonide intervention groups (P 〈0.05).There was a positive correlation between the protein expressions of VEGF and OSM in asthma group (r =0.41,P 〈0.05).[Conclusion]VEGF and OSM are up-regulated in airways and lung tissue of asthmatic mice.And budesonide intervention may lower their expressions.Both are involved in airway remodeling process of asthma.A linear correlation existed between them.Budesonide can affect airway remodeling of asth-ma by down-regulating VEGF and OSM.
