轻揉推拿法对兔骨骼肌急性钝挫伤组织观察及IGFmRNA表达的影响 被引量:2

摘要 目的 探讨轻揉推拿法对骨骼肌钝挫伤修复的作用及其可能机制.方法 将27只实验兔随机分为创伤推拿组(观察组)和创伤对照组(对照组),每组各12只,另3只为正常组.采用"重物自由落体撞击法"制作实验兔骨骼肌急性钝挫伤模型.分别在3d、6d、10d、13d处死实验兔,每组每个时间点处死3只实验兔,每只实验兔在两条后腿致伤腓肠肌部位取标本,共计6个标本,采用光镜组织学观察评分、电镜超微结构观察和应用逆转录聚合酶链式反应(RT-PCR)技术对内源性胰岛素样生长因子(IGF-Ⅰ、IGF-Ⅱ)mRNA的测定,各组数据进行统计学处理.结果 光镜组织学观察评分:创伤推拿组伤后第6、10、13天与创伤对照组比较差异有统计学意义(P<0.05).电镜超微结构观察结果创伤推拿组和创伤对照组在3d未见到肌卫星细胞,6d可见激活的肌卫星细胞,在10d和13d创伤推拿组肌组织趋于正常,创伤对照组有瘢痕形成;RT-PCR技术检测内源性胰岛素样生长因子(IGF-Ⅰ、IGF-Ⅱ)mRNA的表达量:各个时间点内与创伤对照组比较,创伤推拿组内源性的IGF-1和IGF-2的mRNA含量在3、6d与创伤组差异无统计学意义(P>0.05).观察组第10天开始,比创伤组高(P<0.05).结论 轻揉推拿法在中晚期可有效的改善兔骨骼肌钝挫伤后的组织学愈合过程和愈合质量.轻揉推拿法能促进骨骼肌钝挫伤后卫星细胞的激活,进一步促进新肌生成.轻揉推拿法在中晚期对内源性IGF-Ⅰ,IGF-Ⅱ的mRNA表达量上调有促进作用,从而可加强成肌细胞增殖. Objective To study the function of massage in recovery from blunt skeletal muscular contusion and its possible mechanism. Method 27 rabbits were randomly divided into the massage group, the trauma comparison group, each with 12 rabbits, and the normal comparison group with the other 3 rabbits. The model of acute blunt skeletal muscular contusion is made by using the "blow of heavy weight free falling" . Three rabbits from each group were put to death at each time point on the 3rd day, the 6th day, the 10th day and the 13th day, with 2 hind gastrocnemius muscles from each rabbit taken as specimens, 6 specimens in total each time. By the observation and rating based on light microscopic histology, the observation of ultrastructure through electron microscopy, and by using the reverse transcription-polymerase chain reaction ( RT-PCR ) technology to determine the endogenous insulin-like growth factor ( IGF- Ⅰ, IGF- Ⅱ ) mRNA, each set of the data is collected for statistical analysis. Results Based on light microscopic histological rating, it was statistically rewarding to compare the massage group on the 6th day, the 10th day and the 13th day after injury with the trauma comparison group ( P〈0.05 ) . Based on ultrastructural electron microscopic observations, no muscle satellite cells were seen on the 3rd day both in the massage group and the trauma group. On the 6th day, more activated muscle satellite cells were seen. On the 10th chy and the 13th day, the muscle tissue of the massage group tended to be normal, while scars were formed in the trauma group. By using the RT-PCR technology to detect the mP, NA expression levels of endogenous insulin-like growth factors ( IGF- Ⅰ , IGF- Ⅱ ) , a comparison with the trauma group within various time points showed that the massage group had equally the same endogenous IGF-1 and IGF-2 mRNA levels on the 3rd and the 6th day, which has no statistical significance; while from the 10th day onwards, the massage group started to show higher levels than the trauma group ( P〈0.05 ) . Conclusions Firstly, the effects of gentle massage at the intermediate and late stages after the rabbit' s blunt skeletal muscle contusion can effectively improve the histological healing process and healing quality. Secondly, the effects of gentle massage can promote the activation of satellite ceils after blunt skeletal muscle contusion, thus further promoting new muscle formation. Thirdly, the effects of gentle massage at the intermediate and late stages can help increase the endogenous IGF-Ⅰ and IGF-Ⅱ mRNA expression levels, which leads to the reproduction of muscle cells.
