摘要
目的 建立使用促排卵药物后受孕大鼠的子代模型.方法 分别采用20IU、40IU的马血清促性腺激素(PMSG)及人绒毛膜促性腺激素(HCG)对雌性未交配性成熟SD大鼠(7~8周龄)进行超数排卵,并与雄鼠合笼.对照组雌鼠用相同剂量的生理盐水腹腔注射,并与雄鼠合笼.比较其子代出生的时间、个数、体重,分析促排卵模型是否成功;通过平面翻正实验(出生1周时进行)和转动杆实验(出生3周时进行)检测子代大鼠的新生反射和行为能力.结果 各组雌鼠均在合茏后20~22d生产.对照组子代大鼠出生个数(9±1.5)个,体重(8.5±0.4)g.20 IU组子代大鼠出生个数(ll±1.2)个,体重(8.2±0.3)g.40 IU组子代大鼠出生个数(16±4.2)个,体重(7.6±0.25)g.各组大鼠在平面翻正实验和转动杆实验中表现无显著差异(P>0.05).各组雌鼠孕期无显著变化;20IU组与对照组子代大鼠个数之间比较无明显差异性(p>0.05);40IU与对照组子代大鼠个数之间差异有显著性(p<0.01),其效果显著优于20IU组(P<0.05),说明8周龄大鼠促.排卵子代模型的有效剂量为40IU;子代大鼠在新生反射和行为能力方面无显著差异(P>0.05).结论 利用PMSG促排卵后的大鼠,通过产下的子代来作为人类促排卵后受孕后代的模型.
Objective To establish a offspring model after using ovulation induction drugs on pregnant rats. Methods mare serum gonadotropin ( PMSG ) 20IU or 40IU and human chorionic gonadotropin ( HCG ) were injected in female unmated sexual matured SD rats ( 7-8 weeks old ) to superovulated, and then they were mated with male rats. The same dose of saline was given intraperitoneally in control group. Born time, number and weight of offspring were compared to analysis if ovulation model is successful; further experiments including plane righting and pole rotation were performed to detect the reflection and behavioral skills of newborn offspring. Results All female rats delivered in 20-22 days after mates. The number of offspring was ( 9 ± 1.5 ) , ( 11 ± 1.2 ) and ( 16 ± 4.2 ) in control group 201U PMSG group and 40 IU PMSG group separately. Meanwhile in the three group mean weigh was ( 8.2 ± 0.3 ) , ( 7.6 ± 0.25 ) and ( 16 ± 4.2 ) grams separately. There were no differences between the three groups in plane righting and pole rotation. 40IU PMSG group was significantly better than 20IU group ( P〈0.05 ) , and there was significant difference ( P〈0.01 ) between the control group in the number of offspring rats. So 40IU PMSG was an effective dose in ovulation models and there were no difference in neonatal reflection and behavioral capabilities in offspring rats. Conclusion Use of PMSG rats that promote after ovulation, after gave birth to their offspring to stimulate ovulation as a human conception model of offspring
