摘要
目的 探讨酮替芬(KT)抗炎症作用对糖尿病(DM)大鼠的影响及其机制.方法 以高糖高脂饲料对SD大鼠饮食诱导6周,随后一次性给予链脲佐菌素(STZ)制备DM大鼠模型,模型建立后KT实验组给予KT0.09mg/(kg·d)治疗8周,治疗前后分批处死实验大鼠,检测空腹血糖(FBG)、空腹胰岛素(FINS)水平,计算胰岛素敏感指数(ISI)、胰岛素抵抗指数(HOMA-IR),检测甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、炎症因子IL-6和TNF-α.结果 KT可降低FBG、FINS水平(P<0.05),降低TG、LDL-C水平(P<0.05),降低IL-6、TNF-α水平(P<0.05).结论 KT可能通过抑制肥大细胞脱颗粒、抑制NF-κ B激活,改善DM炎症及脂代谢紊乱状态,实现对胰岛细胞的保护作用.
Objective To investigate the effect and mechanism of the anti-inflammatory role of Ketotifen in diabetic rats induced by high- caloriediet and streptozocin. Methods Male SD rats were fed with high-carbon hydrate-fat diet 6 weeks and ip given STZ to in-duce a type 2 diab-etes model. Rats of Ketotifen group were fed with Ketotifen 0.09mg/(kgod), and killed in batches in different time ( o, 8th week ) to test the fasting blood glucose ( FBG ) , and insulin ( FINS ) levels were measured, insulin sensitivity index ( ISI ) and homeostasis model assessment insulin resistance index ( HOMA-IR ) were calculated according to the value of FBG and FINS, to test the level of triglyceride ( TG ) , low density lipoprotein cholesterin ( LDL-C ) , Interleukin-6 ( IL-6 ) and tumor nectosis factor-alpha ( TNF- α) . Results Ketotifen suppressed serum glucose in diabetic rats (P〈0.05) , increase-d ISI and reduced HOMA-IR (P〈0.01) , lowered TG and LDL-C in diabetic rats (P〈0.05) , Ket-otifen reduced the level of IL-6, TNF-α. Conclusion Ketotifen can improve the flammatory and dyslipidaemia state of type 2 diabetic rats by inhibition of mast cell degranulation and NF- κB activation.
