摘要
目的检测结肠癌患者血浆中microRNA-181b(miR-181b)的水平变化,并初步探讨其临床意义。方法采用茎环定量逆转录聚合酶链反应(RT-PCR)方法检测42例结肠癌患者及42例健康人血浆中miR-181b的水平,并分析miR-181b与结肠癌临床病理特征之间的关系。结果与正常对照组(0.38±0.13)相比,结肠癌患者血浆中miR-181b水平(0.64±0.22)明显升高,二者间差异有统计学意义(P<0.01);结肠癌患者血浆中miR-181b水平在不同性别、年龄及肿瘤大小之间差异无统计学意义(P>0.05),而在不同淋巴转移、远处转移及临床分期间的差异具有统计学意义(P<0.05)。结论结肠癌患者血浆中miR-181b水平增高,miR-181b可能参与了结肠癌的发生发展过程。
Objective To investigate the plasma level and clinical significance of microRNA-181b (miR-181b)in patients with colon cancer.Methods The plasma level of miR-181b in 42 patients with colon cancer and 42 healthy controls were detected by stem-loop RT-PCR.The relationship between clinicopathological characteristics of colon cancer and miR-181b was analyzed.Results Compared with healthy controls,the plasma miR-181b in patients with colon cancer was significantly higher (0.38±0.13 vs 0.64±0.22,respectively,P 〈0.01).There was no significant difference between plasma miR-181b and gender,age and tumor size in patients with colon cancer (P 〉0.05).There were significant differences between plasma miR-181b and lymph node metastasis,distant metastasis and tumor stage (P 〈0.05).Conclusion The plasma miR-181b in patients with colon cancer increased and miR-181b may contribute to the development of colon cancer.
