摘要
目的观察巨刺对脑缺血再灌注损伤大鼠脑组织VEGF和Ang-1蛋白表达的影响,探讨巨刺治疗脑缺血再灌注损伤可能的作用机制。方法雄性SD大鼠按随机数字表法随机分为空白组、假手术组、模型组、健侧针刺(即巨刺法)组、患侧针刺组。各组再分为3 d和14 d两个亚组。采用改良线栓法制备脑缺血再灌注损伤大鼠模型。于再灌注24 h后,健侧针刺组针刺人中、百会及电针健侧穴位;患侧针刺组针刺人中、百会及电针患侧穴位。电针选用疏密波,每次持续刺激30 min,每日1次。分别于3 d和14 d光镜下观察大鼠缺血脑组织结构;采用免疫组化法(S-ABC法)测定大脑皮质血管内皮生长因子(VEGF)和血管生成素-1(Ang-1)蛋白表达的变化。结果针刺治疗后大鼠缺血区脑组织损伤程度明显改善,其中健侧针刺组较患侧针刺组改善明显;健侧针刺组、患侧针刺组VEGF及Ang-1的表达均较模型组增多(P<0.05);而健侧针刺组的表达较患侧针刺组显著增多(P<0.05)。结论巨刺能够明显改善再灌注损伤大鼠缺血区脑组织损伤程度,其机制可能与上调脑组织VEGF和Ang-1表达有关。
Objective: To observe the. effects of opposing needling on.the expression of VEGF and, Ang-1 in brain tissue in rats with focal cerebral ischemia-reperfusion injury and explore the' possible, mechanism of.the op- posing needling for treating, rats.with focal cerebral ischemia-reperfusion injury, Methods: One hundred SD made: rats were randomly divided into the blank group,the sham-operated group, the model group, the unaffected-side acupuncture group: (opposing needling group), and the affected-side acupuncture Stoup.with twenty rats in each_ group. And groups were divided into 3d subgroup and 14d subgroup. The model of focal cerebral ischemia-reper- fusion was established by inserting nylon thread into the internal carotid artery. Twenty-four hours after reperfu- sion,Unaffected-side acupuncture group,as well as Affected-side acupuncture group,was treated with “ren- zhong”,“baihui” and electro-acupuncture. Electro-acupuncture was chosen with constant stimulation of the den- sity wave for thirty minutes every day. The ultrastructure of brain tissue on 3 rd and 14 th day after focal cerebral ischemia reperfusion was observed .with the optical microscope. And the expression of VEGF and Angel, in cere, bral cortex tissue was detected with immunehistochemical method. Results: The degree of isehemie .brain tissue damage in rats was improved significantly after acupuncture, But the unaffected-side acupuncture group had an more obvious improvement than that of the affected-side acupuncture group. In comparison with the, sham group the expression of VEGF and Ang-1 in unaffected-side acupuncture group and affected-side acupuncture group increased significantly (P〈 0.05):. And the expression of VEGF and Ang-1 in unaffected-side acupuncture group was mare than that in the .affected-side acupuncture group(P〈 0.05). Conclusion; Opposing needling can greatly improve the injury of ischemia reperfusion of brain tissue in rats ,which may be related to its effect in up regulat- ing VEGF an-d Ang-1 after the ischemia-reperfusion injury.
