摘要
目的推理选育他克莫司高产菌株。方法在研究葡萄糖对他克莫司产生菌影响的基础上,应用葡萄糖的结构类似物2-脱氧-D-葡萄糖对他克莫司产生菌进行抗性选育。结果获得发酵水平较原始出发菌株FCZ0311-7提高19%的突变株7-12,该菌株遗传稳定。结论利用2-脱氧-D-葡萄糖对他克莫司产生菌进行推理选育可行、高效。
OBJECTIVE To obtain tacrolimus high producing strain using rational screen strategy.METHODS Use 2-deoxy-D-glucose,the structural analogue of glucose,to screen high production mutant on the base of studying effect of glucose on the tacrolimus producing strain.RESULTS A high production mutant,strain 7-12,was obtained and the relative fermentation titer of tacrolimus was about 19% higher than that of the original strain,Streptomyces sp.FCZ0311-7.CONCLUSION Rational screen tacrolimus producing strain by 2-deoxy-D-glucose resistance was feasible and efficient.
