不同水体对斑马鱼外周血红细胞微核的影响 被引量:1

摘要 目的通过微核实验检测成都不同采集点的水样对斑马鱼外周血红细胞致突变的影响。方法选取成都市内及周边6个不同地点采集水样饲养斑马鱼,自来水为对照组,饲养至7和14 d时剪尾取血,制备血涂片,显微镜观察并统计微核细胞数,计算微核率,利用SPSS 17.0进行χ2检验。结果斑马鱼无法在毗河水样中存活。其他水样饲养7 d时,与自来水相比,除青城山外其他水样的微核率均显著(P<0.05)或极显著(P<0.01)增高。饲养14 d时,与自来水相比,青城山水样的微核率显著降低(P<0.05),其他水样的微核率均极显著(P<0.01)增高。自来水和青城山水样饲养7和14 d的微核率无显著性差异。其他水样饲养14 d的微核率均显著(P<0.05)或极显著(P<0.01)地高于7 d的微核率。结论除青城山外,其他水样对斑马鱼有致突变性影响,且与暴露的时间成正比,暴露时间越长,致突变率越高。 Objective To test the mutation effect of different water sample on the peripheral red blood cell of zebrafish through micronuclei assay. Methods Zebrafishes were fed in the different water samples collected from 6 different sites of Chengdu and drinking water is the control group. The tails were cut to get blood and prepare blood smear slice after feeding 7 days and 14 days,respectively. Slides were observed using the microscope, and the number and ratio of micronuclei were counted and SPSS 17.0 statistical software was used to analyze data with the chi square test. Results Zebrafishes could not survive in the Pi River sample. Compared with the control, the micronuclei ratio of the water sample except Mountain Qingcheng increased significantly(P〈0.05) or dramatically(P〈0.01) after being fed 7 days. The ratio of Mountain Qingcheng sample decreased significantly(P〈0.05) and the ratio of other water samples increased dramatically(P〈0.01) after being fed 14 days. For drink water and Mountain Qingcheng, there were no significant difference between fed 7 days and 14 days. However, the ratio of the other samples fed 14 days increased significantly(P〈0.05) or dramatically(P〈0.01), compared with that of feeding 7days. Conclusion All different water samples except Mountain Qingcheng had mutation effect on the peripheral blood of Zebrafish. The longer time to expose to them, the higher mutation rate to get.
