摘要
从某生活污水处理厂的活性污泥中筛选出三株对亚甲基蓝具有高效脱色作用的菌株,其中菌株MET-DB2的脱色能力最强。经革兰氏染色,菌株MET-DB2为革兰氏阴性杆菌。考察了培养时间、接种量、染料浓度、振荡速度、p H以及温度对MET-DB2脱色能力的影响。结果表明,MET-DB2对亚甲基蓝脱色的最佳染料初始浓度为150 mg/L,最适温度为30℃,接种量为20 m L,最佳p H为7.50。振荡培养较静置培养条件下的脱色率高且最佳振荡速度为120 r/min。菌株MET-DB2在24 h内对亚甲基蓝的脱色率和去除率分别为89.59%和89.32%。
Three strains, with perfect decolorizing capability for methylene blue, were screened out from the activated sludge from a Sewage treatment plant Among them, the strain MET-DB2 was demonstrated with best decoloring capability. After Gram staining, MET-DB2 was identified as a Gram-negative strain. The paper studied effect of cultivating time, inoculum, the concentration of methylene blue,shaking speed, pH value and temperature on decoloring capability. The results showed that the most preferable initial concentration of the methylene blue was 150 mg/L, the most preferable temperature of MET-DB2 to adsorb the methylene blue was 30℃, the best inoculum was 20 mL, and pH value was 7.50. The decolorization rate at the condition of agitation was higher than stationary condition and the most preferable Shaking speed was 120 r/min. The Decolorization rate and the removal rate can reach to 89.59 % and 89.32 % during 24 hours for methylene blue.
