一株海绵共生真菌ZSDS1-F11次生代谢产物的研究 被引量:3

摘要 对分离自西沙棕色扁海绵Phakelliafusca Thiele的共附生真菌ZSDS1-F11的次生代谢产物进行了研究;运用硅胶柱层析、Sephadex LH-20凝胶柱层析以及半制备高效液相等方法对菌株ZSDS1-F11的发酵产物进行分离纯化;通过理化性质及波谱学方法及参阅文献进行化学结构鉴定;采用环氧合酶-1和环氧合酶-2(cyclooxygenase,COX-1,COX-2)为分子靶标进行炎症抑制剂筛选。结果从菌株ZSDS1-F11的发酵产物中分离得到8个单体化合物。其中化合物1和2具有较弱的环氧合酶-1(cyclooxygenase,COX-1)抑制活性,IC50分别为27.2和2.93μmol。菌株ZSDS1-F11能够产生结构多样性的活性次生代谢产物,并首次报道了化合物1和2具有较弱的抗炎活性。 The secondary metabolites of strain ZSDS1-F11, an endophytic fungus isolated from the spongePhakelliafusca Thiele, were investigated. Fermented products of ZSDS1-F11 were purified by means of chromatography over silica gel column, Sephadex LH-20 and semi-preparative HPLC. Compounds were confirmed based on their physical and chemical properties, spectroscopic analysis and comparison with the relevant literature. Using COX-1 and COX-2 as molecular targets for inflammation inhibitor screening, eight compounds were obtained from the fermented products of strain ZSDS1-F11. Compounds 1 and 2showed weak inhibitory activity against enzyme COX-1, their IC50values were 27.2 and 2.93μmol, respectively. The strain ZSDS1-F11 produced diverse bioactive secondary metabolites. This is the first report on compounds 1 and 2 with weak anti-inflammatory activity.
