肾毒性成分马兜铃酸Ⅰ在木通马兜铃不同部位中的动态累积 被引量:2

摘要 目的测定不同采收期木通马兜铃(Aristolochia manshuriensis Kom.)不同部位中马兜铃酸Ⅰ的含量,考察马兜铃酸I的动态累积规律。方法采用HPLC法,色谱柱为DiamonsilTMC18柱(250 mm×4.6 mm,5μm),流动相为乙腈-体积分数0.05%磷酸水溶液,梯度洗脱,检测波长260 nm,进样量10μL,流速1.0 m L·min-1,柱温30℃。结果从5月中旬到6月中旬,木通马兜铃根和叶中马兜铃酸Ⅰ含量均呈上升趋势,6月中旬叶中马兜铃酸Ⅰ含量达到顶峰后呈现下降趋势,而根中马兜铃酸Ⅰ含量继续呈现上升趋势,至7月下旬根中马兜铃酸Ⅰ含量达到峰值,从7月下旬到9月下旬根中马兜铃酸Ⅰ含量呈现下降趋势,而叶中马兜铃酸Ⅰ含量趋于平稳;从5月中旬到9月下旬,木通马兜铃茎、果实中马兜铃酸Ⅰ含量变化处于平稳趋势。结论木通马兜铃中马兜铃酸Ⅰ的分布具有组织特异性和时效性,为马兜铃酸类化合物生合成途径的研究提供重要的科学依据。 Objective To determine the content of aristolochic acidⅠ( AAI) at different parts of A. manshuriensis collected at different harvest time,and obtain the dynamic accumulation of aristolochic acidⅠ.Methods The content of AAI was determined by RP-HPLC method in which DiamonsilTMC18column( 250 mm × 4. 6 mm,5 μm) was used and the samples was eluted by a gradient elution using acetonitrile and0. 05% phosphoric acid,and the flowrate was 1. 0 m L·min- 1. The temperature of column oven was set to 30 ℃,and the chromatograms were detected under 260 nm. Results The content of AAI at A. manshuriensis leaves and roots was all gradually increased from the middle of M ay to June,and then the content at leaves was gradually decreased to a steady state after it reached the maximum at the middle of June. The peak content of AAI in the roots was reached in late of July and followed with continuous decrease until the middle of September. The content of AAI from A. manshuriensis stems,flowers and fruits were kept constant during the period from the middle of M ay to the late September. Conclusions Distribution of AAI in A. manshuriensis is tissue-and time-dependent,and our finding will provide important scientific basis for the biosynthesis pathway of aristolochic acid compounds.
