摘要
目的:建立HPLC法测定白藜芦醇固体脂质纳米粒(Res-SLN)的载药量与包封率测定方法。方法:采用TopsilC18色谱柱(4.6mm×150mm,5μm),流动相为乙腈-甲醇(25:75),检测波长为306nm,流速为1mL/min,柱温为25℃;采用超滤法分离游离药物与酱藜芦醇固体脂质纳米粒测定包封率与载药量。结果:白藜芦醇在1.01~32.32μg/mL质量浓度范围内与色谱峰面积呈良好线性关系,精密度与回收率均较好;白藜芦醇固体脂质纳米粒平均包封率为81.4%,平均载药量为21.4%。结论:所建方诸准确、可靠、方便,可用于白藜芦醇固体脂质纳米粒包封率与载药量的测定。
Objective :To establishing an HPLC method for determination of resveratrol solid lipid nanoparticle ( Res - SLN) detected rate of drug loading and the coating method. Methods :The Topsil C18 column(4. 6 mm × 150 mm,5 μm) was used,with the mobile phase of acetonitrile - methano (25 : 75 ), detection wavelength 306 nm, flow rate 1 mL/min, column temperature 25 ℃. Results : The calibration curves of the Res was linear in the concentration ranges of 1.01 ~ 32.32μg/mL. The average entrap- ment efficiency of resveratrol solid lipid nanoparticle was 81.4% and the drug -loading rate was 21.4%. Conclusion:The estab- lished method is accurate,reliable and convenient and can be used for the determination of the entrapment efficiency and drug - loading rate of Res - SLN.
