左归丸对绝经后骨质疏松症大鼠骨组织中GPR48、CREB表达影响的实验研究 被引量:13

摘要 目的:通过研究去卵巢骨质疏松症大鼠股骨中GPR48和CREB的蛋白表达,探讨摘除卵巢致肾虚骨质疏松症的病理机制,并研究左归丸的作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制骨质疏松症模型,采用左归丸对实验大鼠治疗12周,以盖天力片作为阳性对照组,正常组、假手术组作为标准对照组,模型组作为空白对照,用ELISA法检测骨质疏松症大鼠股骨GPR48和CREB的蛋白表达。结果:ELISA方法检测表明:正常大鼠股骨中存在GPR48和CREB的蛋白表达。模型组大鼠股骨中GPR48和CREB的蛋白表达水平有明显的下降。左归丸组、盖天力组用药12周后,与模型组相比较,左归丸组可显著上调股骨中GPR48和CREB的蛋白表达水平。结论:1正常大鼠股骨中存在GPR48和CREB的蛋白表达;2股骨中GPR48和CREB的蛋白表达下降可能是导致骨质疏松症发生的机理之一;3左归丸能够上调GPR48和CREB的蛋白表达,表明左归丸对骨质疏松症有一定的防治作用。 Objective : In order to explore the etiological factors and pathology of the osteoporosis due to kidney deficiency, we studied the expressions of GPR48 and CREB in thighbone of the rats of osteoporosis by using ovariectomy to explore its regulating mechanism. Methods : By using ovariectomy, we established the rat model of osteoporosis due to kidney deficiency. We treated the osteoporosis rats with Zuogui Pill for 12 weeks. Simultaneously,we used Gatianli tablets as positive control groups. And rats in the normal group and model group were not given any treatment. We used the ELISA method to investigate the expressions of the GPR48 and CREB in thighbone of the rats. Results : ELISA detection : GPR48 and CREB existed in thighbone. The levels of the ex- pressions of the GPR48 and CREB in thighbone of model group decreased evidently. After the precaution treatment with Zuogui Pill group and Gatianli tablets group for 12 weeks, comparing with the model group, the Zuogui Pill can evidently increase the ex- pression levels of the GPR48 and CREB in thighbone. Conclusion: ①In nomal rats, the thighbone can express the GPR48 and CREB in the protein level. ②The up - regulation of GPR48 and CREB protein expression in the thighbone may be one of the im- portant occurring mechanisms of the osteoperosis due to kidney deficiency. ③The experimental Chinese herbs which have the function of tonifying kidney can increase the level of the expression of the protein of GPR48 and CREB in thighbone. It would pre- vent and treat the osteoporosis due to kidney deficiency.
