摘要
目的 观察Yin Yang 1(YY1)在分化甲状腺癌中的表达,探讨YY1表达与临床特征的关系.方法 使用实时定量聚合酶链反应(Real-time PCR)检测YY1基因在人甲状腺癌细胞株、甲状腺正常/肿瘤组织中的表达;采用Western blot和免疫组织化学染色技术分析YY1蛋白在人甲状腺癌细胞株、甲状腺正常/肿瘤组织中的表达量.结果 在转录水平和蛋白水平,甲状腺癌细胞株中均存在YY1过表达;在甲状腺癌组织中,YY1 mRNA的水平也是明显增加的.此外,免疫组织化学分析表明乳头状甲状腺癌(PTC)和滤泡性甲状腺癌(FTC)中YY1的表达水平明显增加48%和19%.通过YY1蛋白的分层分析,88% PTC患者中出现了YY1的阳性表达.YY1的表达与PTC和FTC患者的临床病理特征的相关性表明,YY1的表达与诊断时的年龄密切相关.结论 YY1过表达于分化甲状腺癌中,且更频繁地过表达于PTC亚型中.
Objective To study the expression of Yin Yang 1 (YY1) in differentiated thyroid cancer,and its correlation with clinical features.Methods We used real-time quantitative polymerase chain reaction (Real-time PCR),Western blotting and immunohistochemical staining techniques to analyze the expression of YY1 mRNA and protein in thyroid cancer cell line,and a series of pairs of normal/ tumor tissues.Results YY1 mRNA and protein over-expression in thyroid cancer cells was detectable.In thyroid carcinoma tissues,YY1 mRNA level was significantly increased.In addition,the immunohistochemical analysis showed that the expression of YY1 in the papillary thyroid carcinoma (PTC) and follicular thyroid cancer (FTC) was increased by 48% and 19%,respectively.The stratified analysis revealed that the positive expression rate of YY1 protein was 88% in PTC.The YY1 expression was closely related to age at diagnosis.Conclusion YY1 is over-expressed in differentiated thyroid cancer,more frequently in PTC subtypes.
