摘要
目的:探讨强制性运动训练对脑缺血大鼠海马区神经细胞存活和c-JNK/p-JNK表达的影响。方法:健康雄性SD大鼠27只,随机分为强制性运动训练组(CIMT)(9只)、对照组(9只)、假手术组(9只),前两组大鼠采用Longa线栓法建立大鼠左侧大脑中动脉缺血模型(MCAO),术中采用激光多普勒血流监测仪监测血流,确保血流下降70%~80%以上。术后第8天开始进行CIMT训练,持续14天。术后第22天采用Western Blot(每组各6只)和免疫组织化学方法 (每组各3只)分别检测患侧海马区c-JNK/p-JNK的表达和新生神经细胞的增生。结果:假手术组患侧海马区存在少量Brd U+/Neu N+细胞,CIMT组患侧海马区Brd U+/Neu N+细胞的数量较对照组有明显的增加(P〈0.05);CIMT组与对照组相比p-JNK表达明显下降(P〈0.05),平衡木评分显著低于对照组(P〈0.05)。结论:CIMT训练可能是通过抑制P-JNK的表达促进大鼠患侧海马区新生神经细胞的存活,促进神经功能的恢复。
Objective To investigate the effect of constraint-induced movement therapy(CIMT)on the survival of newborn nerve cell and expression of JNK/p-JNK in hippocampus of rats with cerebral ischemia.Method Twenty-seven male adult Sprague-Dawley rats were randomly divided into CIMT group, control group and sham operation group.Rats in the CIMT and control groups were subjected to left middle cerebral artery occlusion(MCAO),ensuring the decrease in blood flow by at least 70% to 80% through dynamically monitored by Laser Doppler Flowmetry. CIMT was performed 8 days after MCAO and lasted for 14 days.Western blot and double-label fluorescence method were respectively used to detect the JNK/p-JNK expression and newborn nerve cells proliferation. Results There was a few Brd U+/ Neu N+cells in the sham group, and more Brd U+/ Neu N+cells appeared in the CIMT group as compared with the control group(P〈0.05). As compared with the control group,the p-JNK expression increased significantly(P〈0.05),the neural function recovered well(P〈0.05) in CIMT group. Conclusion Constraint-induced movement therapy could obviously promote the survival rate of the nerve cells in hippocampus and the recovery of neural function in rats with cerebral ischemia through inhibiting the p-JNK expression levels.
