摘要 目的分析血管紧张素Ⅱ(AngⅡ)对人血管内皮细胞和猪血管内皮细胞的活性氧和线粒体功能的影响。方法体外培养血管内皮细胞ECV-304和PAE,用不同浓度AngⅡ处理细胞,采用流式细胞术分析细胞内活性氧、线粒体膜电位和细胞凋亡。结果血管内皮细胞内的ROS水平随AngⅡ浓度增加而增加,AngⅡ导致线粒体功能损伤和凋亡。结论血管紧张素以浓度依赖的方式导致内皮细胞活性氧水平增加,并参与血管内皮细胞的损伤。 Objective To investigate the effect of AngiotensinⅡ( AngⅡ) on reactive oxygen species and mitochon-drial function in Human vascular endothelial cells and pig vascular endothelial cells .Methods After ECV-304 and PAE cells were treated with different concentrations of AngⅡ for different durations , reactive oxygen species , mito-chondrial membrane potential and cell apoptosis were detected by flow cytometry .Results AngⅡinduced apoptosis of ECV-304 and PAE cells.Treatment with AngⅡreduced cell membrane potential .Moreover,AngⅡtreatment signifi-cantly increased the level of reactive oxygen species in ECV-304 and PAE cells.Conclusion AngⅡsignificantly in-duces apoptosis of vascular endothelial cells possibly by increasing the reactive oxygen species level .
