摘要
目的通过研究转移抑制基因1(MTSS1)在人肺鳞癌组织中的表达,探讨MTSS1在肺鳞癌中的临床意义。方法收集2005年9月至2009年12月病理诊断明确,手术切除的80例肺鳞癌标本,采用免疫组化染色法检测MTSS1蛋白在肺鳞癌中的表达,探讨MTSS1与肺鳞癌临床病理特征及其预后的关系。另取10例正常肺组织标本作对照。结果 (1)MTSS1蛋白主要表达于细胞浆,在正常肺组织中阳性表达率(90.0%)明显高于肺鳞癌中的阳性表达率(53.8%)(P<0.05)。(2)随着组织分化程度由高到低,MTSS1的阳性表达水平呈现由高到低的变化;无淋巴结转移的病例中MTSS1的阳性表达明显高于出现淋巴结转移者(P<0.01)。(3)Kaplan-Meier生存分析显示,MTSS1表达阴性或缺失的肺鳞癌患者的预后明显较MTSS1表达阳性患者差(P<0.01);Cox多因素生存分析显示,影响患者预后的独立危险因素为MTSS1无表达和低表达(P<0.01)、肿瘤组织分化程度低(P<0.05)、临床分期高(P<0.05)、肿瘤直径大(P<0.05)、有吸烟史(P<0.01)及淋巴结转移(P<0.01)。结论 MTSS1的表达水平可以作为一项新的独立指标,用以判断肺鳞癌患者的预后。
Objective To investigate the expression of metastasis suppressor 1 ( MTSS1 ) protein in the tissues of human lung squamous cell carcinoma and its clinical significance. Methods Eighty specimens of surgical excision with distinct pathological diagnosis of lung squamous cell carcinoma were collected from November 2005 to Deeemner 2009. The expres- sion of MTSS1 protein in tissues of lung squamous cell carcinoma was detected by immunohistochemistry, and the relation- ship between MTSS1 and clinicopathologieal features, prognosis of lung squamous cell carcinoma was explored. In addition, 10 specimens of normal lung tissues were selected as control. Results (1) The MTSS1 protein mainly expressed in cyto- plasm, and its expression rate in normal lung tissue was significantly higher than that in squamous cell carcinoma tissues (90. 0% vs 53.8% ,P 〈 0.05 ). (2)The levels of MTSS1 positive expression presented the changes from high to low with the tissues' differentiation degrees from high to low. The MTSS1 positive expression rate in patients without lymph node me- tastasis was significantly higher than that in patients with lymph node metastasis (P 〈 0. 01 ). (3) Kaplan-Meier survival a- nalysis showed that the patients with negative or absent MTSS1 expressions had poor prognosis compared with the patients with positive MTSS1 expressions. The Cox multivariate survival analysis showed that the independent risk factors influencing prognosis of patients were associated with negative or absent expressions of MTSS1 level (P 〈 0.01 ) , differentiation degree of tumor tissues ( P 〈 0.05 ), clinical stage ( P 〈 0.05 ), tumor of large size ( P 〈 0.05 ), smoking history ( P 〈 0.01 ) and lymph node metastasis ( P 〈 0.01 ). Conclusions The expression level of MTSS1 can be used as a new independent index for judging the prognosis of lung squamous cell carcinoma patients.
