摘要
通过优化发酵培养基中的成分及其含量,提高诺沃霉素有效组分的产量。以高效液相色谱检测目标产物的峰面积作为指标,通过单因素试验获得最佳碳源及氮源。使用正交试验等获得最佳发酵培养基:葡萄糖4.0%,酵母浸粉0.5%,豆粕1.0%,磷酸氢二钾0.035%,碳酸钙0.05%,豆油0.1%。相关发酵条件为p H 7.0,种龄96 h,接种量10%,发酵温度28℃,发酵时间120 h。根据高效液相色谱检测结果,在最佳发酵条件下,目标产物产量比对照培养基提高了657%。
In order to increase the production of Novonestmycin produced by Streptomyces phytohabitans HBERC-20821,the composition and content of the fermentation media were optimized using single-factor test and orthogonal design. According to the result of HPLC, the optimum parameters were obtained as follows: 4.0% glucose, 0.5% yeast extract, 1.0% soybean meal, 0.035% K2HPO4,0.05% Ca CO3, 0.1% soybean oil. p H 7.0,seed age 96 h, inoculum volume 10%, fermentation temperature 28 ℃, fermentation time 120 h. Under the optimized conditions,the product output of Novonestmycin increased by 657%compared with the contrast medium.
