摘要
以带腋芽的广西柳州野生葡萄茎段为外植体,研究0.1%Hg Cl2不同灭菌时间、75%酒精和0.1%Hg Cl2两种灭菌剂组合对其灭菌效果的影响;同一灭菌处理对其不同茎段部位灭菌效果的影响;不同取材季节同一灭菌处理对其灭菌效果的影响。采用同一培养基(MS+6BA 1.0 mg/L+NAA 0.1 mg/L+糖30 g/L+琼脂6 g/L)对灭菌后的外植体进行培养。试验结果表明:0.1%Hg Cl2灭菌时间越长,对野生葡萄外植体的灭菌效果越好,但出芽率则随灭菌时间增加呈先上升达最大值后逐渐下降的趋势;两种灭菌剂组合随75%酒精灭菌处理时间越长灭菌效果越好,但出芽率则随灭菌时间增加呈先上升达最大值后再下降的趋势;在同一灭菌处理下,以茎段上端部位的灭菌效果最好,出芽率最高;在春季取材,灭菌效果最佳,出芽率最高。
The stems with axillary bud of wild grape in Guangxi Liuzhou were used as the explants and the effects of different sterilization time with 0.1% HgCl2, different sterilization combinations with 75%acohol and 0.1% HgCI2, different stem positions and sample seasons were studied . The explants after sterilization were cultured on the same medium MS+6-BA 1.0mg/L+NAA 0.1mg/L+Sugar30g/L+Ag6g/L.The experiment results indicated that the longer sterilization time, the better sterilization effect, in which the bud induction rate showed the tendency that go up first then go down with the sterilization time increasing. With the same sterilization treatment, the upper stem showed the best sterilization effects and the highest bud induction rate .Sample in spring could get best sterilization effects and the highest bud induction.
