大岩桐试管花芽分化与开花影响因素 被引量:2

摘要 以大岩桐(Sinningia speciosa)试管苗为试验材料,研究了基本培养基种类、植物生长调节剂种类、培养基不同成分、赤霉素质量浓度及植株营养生长状态对诱导试管内花芽分化的影响。结果表明,WPM培养基是适宜大岩桐诱导花芽分化的基本培养基种类;适宜的细胞分裂素6-BA与生长素NAA质量浓度配比,对提高大岩桐花芽分化率较为有利;诱导大岩桐试管内花芽分化的适宜培养基为:WPM+6-BA0.3 mg·L-1+NAA0.2 mg·L-1+蔗糖40 g·L-1+琼脂9 g·L-1;赤霉素不利于健壮生长。3 cm的苗径大小是大岩桐营养生长向生殖生长转化的临界条件,达到临界条件后,花芽分化率随苗径增加而逐渐增多。 With aseptic plantlets of Sinningia speciosa, we studied the effects of basic mediums, plant growth regulators, differ- ent concentrations of mineral, 6-BA, NAA, sugar and agar, concentrations of gibberellin, and plantlet physiological sta- tus on in vitro flowering. The most suitable basic mediums was WPM, while the combination of cytokinin 6-BA and auxin NAA was suitable for flower bud differentiation of S. speciose. WPM medium with 6-BA 0.3 mg· L^-1 , NAA 0.2 mg ·L^-1 , sugar 40 g · L^-1 and agar 9 g· L^-1 was suitable for inducing in vitro flower buds differentiation. Gibberellin made against with vigorous growth. The 3-cm seedling diameter of the plantlets was the critical condition of the transition from vegetative to reproductive growth for S. speciosa, and with the increase of the plantlet seedling diameter, the percentage of floral bud differentiation was increased.
