摘要
为探讨"大通犊牦牛"组织线粒体COX活性与亚基COX-Ⅰ、ⅡmRNA表达量之间的关系。采用酶联免疫分析法(ELISA)对犊牦牛心肌、骨骼肌、大脑、肝脏组织线粒体COX活性进行测定;绝对定量荧光PCR(Real-time PCR)检测组织中COX-Ⅰ、ⅡmRNA表达量。结果显示:大通犊牦牛心肌、骨骼肌、大脑、肝脏组织线粒体COX活性高于乐都犊牦牛;大通犊牦牛组织间COX-Ⅰ基因拷贝数明显高于乐都犊牦牛(P<0.05);大通犊牦牛组织间COXⅡ拷贝数以及乐都犊牦牛COXⅡ拷贝数均差异显著(P<0.05),大通犊牦牛不同组织中的COX-Ⅰ、Ⅱ的表达量与乐都犊牦牛相比差异显著(P<0.05)。结果表明:含野血的大通犊牦牛组织线粒体COX的氧化磷酸化作用要强于家犊牦牛;线粒体COX两种亚基mRNA变化与组织和品种有关;COX活性变化可使COX-Ⅰ、ⅡmRNA发生失调。
To study the relationship between tissue mitochondrial COX activity and subunitCOX- Ⅰ,ⅡmRNA expression in Datong yak calf. Used ELISA to measure the tissue COX activity of calf cardiac, skeletal muscle, brain and liver. Used Real time PCR to detect the expression of COX- Ⅰ,Ⅱ mRNA in tissue. In Da'tong yak calf myocardium, skeletal muscle, brain, liver, mithchondrial COX activity was higher than that of Ledu yak. Copy number of COX- Ⅰ gene in Datong yak calf tissue was significantly higher than that of Ledu yak calves (P〈0.05). Copies number of COX- Ⅱ gene in Datong yak calf tissue and COX-Ⅱ of Ledu yak calves showed a significant differences (P〈0.05). Expression in different tissues COX-Ⅰ, Ⅱbetween Datong yak calf and the Ledu calves yak had significant differences (P〈0.05). Phosphorylation of mitochondrial COX at Datong yak calf with wild blood was higher than that of Ledu yak calves. Two subnits of mitochondrial COX varied with tissues and breed. The changes of activity of COX can make the COX- Ⅰ,ⅡmR NA expression disorders.
