促血管生成素-1对糖尿病大鼠CD133表达的影响及相关性研究

摘要 目的探讨促血管生成素-1(Angiopoietin-1,Ang-1)对糖尿病大鼠肾组织CD133的影响及CD133表达变化与糖尿病的相关性。方法将SD大鼠分为正常对照组(NC)、糖尿病肾病组(DN)、Ang-1干预组(AV)及空病毒组(BV)。采用STZ腹腔注射诱导糖尿病模型。成模8周后尾静脉注射Ang-1腺病毒载体。多时点检测24h尿蛋白、血糖及肾组织CD133蛋白表达水平。结果DN、AV、BV组尿蛋白、血糖、肌酐均较NC组升高,肾组织CDl33表达增强,差异有统计学意义(P〈0.05),其中AV组CD133表达增强最明显,峰值为20—28w,20w后AV组尿蛋白和血糖较DN及BV组升高变缓(P〈0.05)。24h尿蛋白排泄量和血糖水平分别与CD133表达水平呈正相关(P=0.001,r=0.617)和负相关(P〈0.01,r=-O.610),Ang-2免疫组化IOD表达水平与CDl33免疫组化IOD表达水平呈正相关(P〈0.01,r=0.622),Ki67免疫组化IOD表达水平与CD133免疫组化IOD表达水平呈正相关(P〈0.01,r=0.624)。结论Ang-1能促进糖尿病大鼠肾脏微小血管内皮层CD133的表达,降低血管渗漏和尿蛋白排泄。 Objective To explore the effect of Ang-1 on the expression of CD133 in diabetic rats and its correlation. Methods Rat model of diabetic nephropathy was induced by a single intraperitioneal injection of STZ. The subjects were divid- ed into four groups:control group (NC), diabetic nephropathy group (DN),Ang-l-vector group (AV) and blank vector group (BV). The urinary protein, blood glucose and the expression of CD133 were observed at 8th, 12th,20th and 28th week. Re- suits 24h UAlb, blood glucose and creatinine in DN,AV, BV groups were higher than those in NC group,but increased 24hUAlb and blood glucose in AV group decreased after 20 weeks (/9〈0.05). Immunostaining score of CD133 level in DN, AV, BV groups increased compared with NC group (P〈0.05), particularly in AV group with peak levels from 20 to 28 weeks. The level of uri- nary protein was positively correlated with the expression of CD133 (P=-0.001 ,r=-0.617). The expression of CD133 was negatively correlated with the level of blood glucose (P〈0.01 ,r=-0.610), positively correlated with the expression of Ang-2(P〈0.01 ,r= 0.622), positively with the expression of Ki67 (P〈0.01 ,r=0.624). Conclusion Ang-1 can increase the expression of CD133 in diabetic rats, reduce the vascular leakage and urinary albumin excretion.
