摘要
[目的]对沙门氏菌和金黄色葡萄球菌不同的DNA提取方法进行评价。[方法]采取3种不同的方法对沙门氏菌和金黄色葡萄球菌的DNA进行提取,利用全波长紫外/可见光扫描分光光度计比较其浓度和纯度。[结果]方法 3提取的DNA质量最好,浓度和纯度都较高,方法 1提取的DNA质量次之,纯度较高,浓度次之,方法 2提取的DNA浓度一般,但存在碳水化合物或盐类污染。[结论]方法 3是一种适用于沙门氏菌和金黄色葡萄球菌以及其他病原菌的有效提取方法。
[ Objective] To evaluate different extraction methods used for Salmonella and Staphylococcus aureus genome DNA. [ Method] Three kinds of methods have been applied to extract the DNA from Salmonella and Staphylococcus aureus. The concentration and purity were compared using full wavelength ultraviolet/visible light scanning spectrophotometer. [ Result] The quality of DNA extrated by using method 3 was superior to the other methods, also, he concentration and purity of DNA was relatively high. As to the DNA extracted by method 1, its purity was no less than which extracted by method 3 while concentration is lower. So the quality of DNA extracted by using method 1 was estimated to be second only to method 3. Extrating DNA by using method 2 seemed fine, but the fouling of carbohydrates or salts has come into being a question. [ Conclusion ] Method 3 of this study is effective and generally applicable to Salmonella, Staphylococcus aureus and other pathogenic bacteria for DNA extraction.
