摘要
目的 改进并提高青阳参的质量控标准。方法 针对原标准存在的部分方法繁锁、专属性不强,并使用危害较大的试剂,缺乏含量测定和显微鉴别等问题,建立青阳参显微、TLC鉴别方法以及青阳参中青阳参苷元的HPLC含量测定方法,以改进和提高质量标准,采用Omni Gene LLC生厂的HPLC Column Hubble C18(5μm 150×4.6 mm)色谱柱;以乙腈-0.2%磷酸溶液(24:76)为流动相;检测波长为259 nm;流速为1.0 ml/min。结果 改进后的显微特征专属性强,TLC能检出青阳参对照药材和青阳参苷元对照品,无其他干扰,建立的含量测定HPLC条件下,青阳参苷元在10.044-200.880μg/ml浓度范围内线性关系良好,r=0.999 989;平均回收率为95.68%,RSD=2.84%(n=9)。结论 改进提高后的方法简便、准确、重现性好,能够更有效控制青阳参的内在质量。
Objective To study on quality control of qingyangshen. Methods Qingyangshen chinensis were identified by microscopic characteristic and determination of cynanchagenin in qingyangshen by HPLC. Column was column hubble C18(5 μm 150×4.6 mm)and with acetonitrile-0.2 phosphoric(24:76)as mobile phase. The detection wavelength was at 259 nm. The flow rate of 1.0 ml/min. Results Microscopic characteristic specificity was strong,accurately cynanchagenin were identified,a linear relationship of cynanchagenin was good within 10.044 μg/ml - 200.880μg/ml(r=0.999 989). The average recovery was 95.68%with RSD 2.84%(n=9). Conclusion The method is simple,accurate,reproducible and can be used for the quality control of qingyangshen.
