摘要
目的观察马齿苋总黄酮(Portulaca oleracea total flavones,POTF)对异丙肾上腺素(isoproteronol,ISO)致小鼠实验性心肌缺血的保护作用,并探讨其可能的作用机制。方法 60只昆明种小鼠随机分为6组,即正常对照组、缺血模型组、POTF给药组(80、40、20 mg/kg)、普萘洛尔给药组(5 mg/kg);采用腹腔注射ISO诱导小鼠造成心肌缺血的动物模型,记录小鼠标准Ⅱ导联心电图ST段(J点)上移值及心肌含水量(MWC)、心肌指数(MI)的变化,观察小鼠常压密闭条件下的存活时间;检测血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)、丙二醛(MDA)含量和心肌组织中超氧化物歧化酶(SOD)、MDA、Ca2+-ATPase活性变化。结果 POTF给药组能明显降低J点上移值;延长心肌缺血小鼠常压缺氧存活时间(P<0.05);降低心肌缺血小鼠血清中LDH、CK、MDA的含量(P<0.05);升高心肌组织SOD、Ca2+-ATPase活性,降低MDA含量(P<0.05);明显减少MWC、MI。结论 POTF对小鼠急性心肌缺血损伤具有保护作用,其作用可能与增强抗氧化能力、抑制脂质过氧化、提高耐缺氧能力有关。
Objective To explore protective effect of Portulaca oleracea total flavone (POTF) on acute myocardial ischemia injury induced by isoproterenol(ISO)in mice and its possible mechanism. Methods Kunruing mice were randomly divided into 6 groups, the normal control group, ischemia model group, POTF to drug group (80, 40, 20 mg/kg), the propranolol drug group (5 mg/kg). Myocardial ischemia model was made by intraperitoneal injection of ISO, the extent of myocardial damage was evaluated with ST segment in lend II on ECG, myocardialwater content ( MWC ) and myocardial index ( MI ), and the survival time the mice model under the hypoxic condition was recorded;the survival time of mice under the condition of normal pressure airtight was observed. Content of creatine kinase (CK) lactate dehydrogenase ( LDH) and content of malondialdehyde (MDA) , in serum and activity of the super oxide dismutase ( SOD ) , content of MDA and calcium ion (Ca2+) in myocardial tissue homogenate were detected. Results The ST segment displacemerit in lend II on ECG was improved in POTF group, and it extended the survival time of normal pressure hypoxia ischemia mice, and the content of serum CK and LDH decreased significantly in POTF group compared with those in the model group, activity of SOD and Ca2+-ATPase increased, content of MDA, significantly reduce the MWC, MI in POTF group. Conclusion POTF plays a protective role in myocardial ischemia in mice, and the protective effect mechanism may be related to its inhibition of the oxygen free radical and enhance oxidation resistance.
