补肾化痰方对多囊卵巢综合征大鼠性激素及卵巢形态学的影响 被引量:7

Effects of Bushen Huatan Fang on Sex Hormone Levels and Ovarian Morphology in Rats with Polycystic Ovarian Syndrome

摘要 目的:观察补肾化痰方对多囊卵巢综合征(polycystic ovarian syndrome,PCOS)大鼠性激素及卵巢形态学的影响。方法:采用皮下注射脱氢表雄酮造模法建立PCOS大鼠模型,60只造模大鼠随机分为模型对照组、二甲双胍组、补肾化痰方低剂量组、补肾化痰方高剂量组,另取15只大鼠为正常对照组,各组给予相应药物连续灌胃28 d。行阴道涂片、HE染色观察动情周期;用酶联免疫吸附测定法测定血清促黄体生成素、睾酮水平;行卵巢组织HE染色,光镜下观察卵巢组织形态学变化。结果:与对照组比较,模型组大鼠无规律的动情周期,血清促黄体生成素、睾酮水平升高,卵巢表面苍白、体积增大、黄体数目减少、呈多囊样改变。二甲双胍和补肾化痰方两个剂量组可明显降低模型大鼠血清促黄体生成素、睾酮水平,恢复大鼠动情周期,增加各级卵泡数目及黄体数目,改善卵巢形态学结构。与补肾化痰方低剂量组比较,补肾化痰方高剂量组降低睾酮水平更明显。结论:补肾化痰方可从总体上调节下丘脑-垂体-卵巢轴功能,降低多囊卵巢综合征大鼠促黄体生成素、睾酮水平,恢复大鼠动情周期,改善卵巢形态学结构。 Objective:To observe the effects of Bushen Huatan Fang on sex hormone levels and ovarian morphology in rats with polycystic ovarian syndrome. Methods:We established the model of PCOS rats using subcutaneous injection of dhea mode method. 60 Rats were randomly divided into 5 groups:model control group,Metformin group,Bushen Huatan Fang of low dose group and high dose group,an-other 15 rats as normal control group. Rats in all groups were treated with corresponding drugs by gastrogavage for 28 days. Observed es-trous cycles by vaginal smear and HE staining methods and detected luteinizing hormone(LH)and testosterone by ELISA. Observed o-varian morphology by HE staining and light microscopy. Results:Compared with control group,rats in model group had higher LH and testosterone levels,having irregular estrous cycle,multiple follicular cysts and less corpus luteum. Metformin and Bushen Huatan Fang could improve the pathological changes of PCOS rats above at different degree. Bushen Huatan Fang had better effect on decreasing tes-tosterone level in high dose group than in low dose group. Conclusion:Bushen Huatan Fang could reduce LH and testosterone levels,re-store estrus cycle and improve ovarian morphology in PCOS rats.
