摘要
目的:探讨和比较穴位注射、穴位埋线保护胰岛β细胞功能的机制。方法:将50只雄性SD大鼠随机分成空白对照组、模型组、穴位注射组、穴位埋线组。空白对照组用普通饲料喂养,其余3组用高糖高脂饲料喂养4周,并按体质量35 mg·kg-1右侧腹腔注射链脲佐菌素复制2型糖尿病大鼠动物模型。空白组注射等量柠檬酸缓冲液,穴位埋线组予双侧足三里、三阴交、胃脘下俞埋线,穴位注射组在相同穴位注射参麦注射液,均隔日1次,共4周。观察各组大鼠的血糖水平、胰岛形态学变化及pander、Caspase-3蛋白的表达。结果:穴位埋线组、穴位注射组空腹血糖显著低于模型组(P<0.05),穴位注射组下降更显著。穴位埋线和穴位注射组大鼠胰腺组织内的胰岛细胞损伤少于模型组(P<0.05),穴位注射组胰岛细胞损伤更少。与空白组比较,模型组胰腺pander、Caspase-3蛋白表达上升(P<0.05);穴位埋线组与穴位注射组胰腺pander蛋白表达较模型组下降(P<0.05),穴位埋线组下降更显著。结论:穴位注射、穴位埋线或许通过降低血糖,抑制高血糖对胰岛β细胞pander的激活和Caspase-3表达,以减少胰岛β细胞凋亡,缓解2型糖尿病的进程,且穴位注射改善胰岛β细胞功能更显著。
Objective:To study the mechanism of acupoint injection and acupoint catgut embedding in protecting islet beta cell function. Methods:50 male SD rats were randomly divided into blank control group,model group,acupoint injection group and the acupoint catgut embedding group. The blank control group was fed with normal diet,the other three groups with high fat and high sugar diet for 4 weeks, and according to the weight of the 35mg·kg - 1 by intraperitoneal injection of STZ induced type 2 diabetic rats. The blank control group was injected with citrate buffer solution,acupoint catgut embedding group,bilateral Zusanli( ST36),Sanyinjiao(SP6),Weiwanxiashu (EX-B3)catgut embedding,acupoint injection of Shenmai injection in acupoint injection were the same,once every other day,a total of 4 weeks. Observed the level of blood glucose,islet morphology and pander,Caspase-3 protein. Results:The fasting blood glucose of acu-point catgut embedding therapy group and acupuncture point injection group was significantly lower than that of model group( P ﹤0. 05),acupoint injection group decreased more significantly. The acupoint catgut embedding therapy group and acupuncture point in-jection group in pancreas islet cell damage was less than the model group(P ﹤ 0. 05),acupoint injection group islet cells even less. Compared with the blank group,model group increased expression of pander and Caspase-3 protein expression in the pancreas;acupoint catgut embedding group and acupoint injection group pander protein in pancreas decreased compared with the model control group,acu-puncture point injection group decreased more significantly. Conclusion:Acupoint injection and acupoint catgut embedding therapy may inhibit high glucose on islet beta cells of pander activation and Caspase-3 expression by lowering blood glucose,reduce the apoptosis of islet beta cell,alleviate the progression to type 2 diabetes,and acupoint injection improved the function of islet beta cells is more signifi- cant.
