摘要
目的:观察自发性高血压大鼠血脂及房室结(AVN)形态结构变化,探讨厄贝沙坦对其影响。方法:将WKY雄性大鼠和SHR鼠随机分为:1WKY组(对照组)10只;2SHR组(模型组)10只;3厄贝沙坦治疗组(治疗组)10只(厄贝沙坦0.6 m L·100 g-1·d-1灌胃)。WKY组和SHR组同等剂量的生理盐水每天灌胃1次,持续喂养8周。测定血脂变化及光镜下观察房室结病理形态学变化。结果:治疗8周后:(1)血脂变化:与对照组比较,SHR组大鼠血清TC、TG、LDL均显著升高,而HDL显著降低(P<0.05);与SHR组比较,治疗组大鼠血清TC、TG、LDL均显著降低,而HDL显著升高(P<0.05)。(2)对照组大鼠房室结区域内细胞小而密,核正常,房室结结细胞无变性、坏死。SHR组房室结结细胞排列紊乱,断裂,胞核浓缩深染,可见少量的炎症细胞侵入。治疗组房室结结细胞排列趋于整齐,与SHR组比较明显减轻。结论:SHR大鼠房室结8周后出现间质胶原重塑;厄贝沙坦能降低SHR大鼠的血压,改善心功能,可不同程度下调房室结间质胶原的沉积。
Objective: To investigate the changes of the morphology and structure of atrioventricular node and the intervention role of irbesartan in spontaneously hypertensive rats. Methods: WKY and spontaneously hypertensive rats were randomly divided into three groups with 10 rats in each group: 1WKY group( control group); 2SHR group( the model group); 3irbesartan treated group( the treatment group). The treated group was injected with irbesartan 0.6 m L·100 g- 1·d- 1and performed intragastric administration for 8 weeks,while SHR group and WKY group were treated with the same dose saline. Then the changes of blood lipid and the pathological changes of atrioventricular node were observed under light microscope. Results: After 8 weeks of treatment,blood lipid changes in the model group were significantly higher than that in the normal group,while the HDL values were significantly decreased( P〈 0. 05) in the model group. Compared with the model group,the serum TG,LDL and TC in the treatment group were significantly decreased,while the HDL value was significantly increased( P 〈0. 05). In the control group,the cells were small and dense in the atrioventricular node area,but there was no degeneration and necrosis of the atrioventricular node cells. In the model group,the arrangement of the atrioventricular node cells was disordered; the fracture was concentrated in the cytoplasm,and there was a small amount of inflammatory cells in the model group. In the treatment group,the arrangement of atrioventricular node cells tended to be neat,and the model group was significantly reduced. Conclusion: Eight weeks later,the AVN of model group of rats,appears interstitial collagen remodeling. Irbesartan can reduce the blood pressure of the rats in the model group and improve cardiac function. The deposition of interstitial collagen in the atrioventricular node can be reduced to varying degrees.
