摘要
谷胱甘肽硫转移酶(GSTs)受黄萎病菌诱导表达,对黄萎病具有抗性。为明确该基因在棉花中抗黄萎病功能,本研究在前期工作基础上,以陆地棉Coker312为受体,利用农杆菌介导的茎尖转化法对GhGST基因进行遗传转化,共获得132株卡那霉素抗性植株,利用PCR检测以及胶回收测序进一步确定了11株稳定的转基因阳性棉植株。转基因植株的获得为后续GhGST基因功能研究及棉花新品种培育提供了基础材料。
Glutathione S‐transferase (GSTs) is induced by verticillium wilt and has resistance to verticillium wilt .In order to investigate the function of the gene in cotton resistance to ver‐ticillium wilt .In this paper ,the transgenic plants with GhGST were obtained based on shoot tip‐agrobacterium medium using Coker312 as the receptor .Through this method ,132 Kana‐mycin (Kan) resistant plants were obtained from Coker312 shoot tips .From them ,11 plants with the GhGST were transgenic according to the PCR and sequencing method testing .These plants can be used as the basic germplasm of GhGST function analysis and cotton breeding in follow‐up research .
