摘要
为明确抗叶锈病相关基因在抗叶锈性中的作用,本研究以小麦叶锈菌诱导的TcLr19为研究对象,利用病毒介导的基因沉默(Virus induced gene silencing,VIGS)技术对前期克隆得的TaSTKC8基因进行了初步的功能分析,结果显示TaSTKC8部分基因沉默后,反应型由"0"变为"X";组织学观察显示接种叶锈菌120h,寄主组织中虽然出现了坏死反应,但有大量菌丝的生长,说明TaSTKC8基因参与了TcLr19对小麦叶锈菌THTS的抗病反应。
The objective of this research was to study the disease‐resistant mechanism of wheat leaf rust resistance‐associated gene .The functional analysis of the resistance related the gene TaSTKC8 cloned from TcLr19 was carried out using BSMV‐VIGS technique .The infection type of the control wheat TcLr19 was“0” ,while the wheat plants carrying the silenced gene TaSTKC8 with VIGS showed infection type of “X” .Further histological observation indica‐ted the same results .Plenty of mycelia were observed in the plant with silenced gene TaST‐K C8 at 120 hours post‐inoculation .This result indicates that the product of gene TaS TK C8 is required for Lr19‐mediated resistance to race 09‐12‐284‐1 (THTS) .
