摘要
以长白猪(Landrace)cDNA为模板,克隆STAT1基因cDNA全长.家猪STAT1基因cDNA全长编码757个氨基酸的前体蛋白,与人、大鼠、小鼠、牛等哺乳动物的STAT1氨基酸序列一致性分别为95%、90.8%、89.6%、91.6%;同时,本研究亦鉴定到家猪STAT1的3种剪接变体(分别命名为STAT1-sv1,STAT1-sv2,和STAT1-sv3).采用RT-PCR方法,进一步检测了STAT1及其3种剪接变体在成年家猪各组织中的表达情况.STAT1和两种剪切变体(STAT1-sv2和STAT1-sv3)在家猪组织中广泛表达,而剪接变体STAT1-sv1则在肺和脾脏中有较高表达水平.
In this study, the full-length cDNAs of pig STAT1 (pSTAT1) gene was cloned using RT-PCR. The cloned pSTAT1 encodes a pro-protein of 757 amino acids, which shares high amino acid se-quence identity with that of human (95%), mouse (89.6%), rat (90.8%) and cattle (91.6G). In ad-dition, 3 novel mRNA splice variants of STAT1, named pSTATI-svl, pSTATI-sv2 and pSTATI-sv3respectively, were identified in this study . RT -PCR assay showed that STAT1 and 2 splice variants(pSTATI-sv2 and pSTATI-sv3) are expressed in all tissues examined, while pSTATI-svl is abundantlyexpressed in spleen and Lung. These findings pave the way for further elucidation of the roles of STAT1gene in pigs.
